Hericium erinaceus is a well known edible and medicinal fungus which is attached to the basidiomycetes phylum and Hericium family.It has been reported that H.erinaceus contains varieties of beneficial granulose,essential amino acids and microelements.It has been used traditionally in china to treat gastric ulcer,chronic gastritis and other digestive tract related diseases.Many active substances about H.erinaceus have been reported.But most of those were about polysaccharides.The studies of oligosaccharides from H.erinaceus, especially from the fruiting body of H.erinaceus were scarce.Oligosaccharide is a kind of chemical compound which is polymerized with 2 to 10 monosaccharides.It has variety of bioactivities including improving lipids,hypoglycemic,antioxidation and improving gastrointestinal tract,etc.It is widely used in the food,feeder, pharmaceutics and cosmetics industries.The manufacture of the oligosaccharide is mainly through the catalytic action of transferase or hydrolyase now.There were not much of the reports about the extraction of oligosaccharides from plants.And the studies of oligosaccharides from edible fungus were limited.The method of grading-alcoholic precipitation was firstly used to separate the extraction from the fruiting bodies of H.erinaceus in this paper.And then we detected the reducing sugar of the extractions with the reagent of DNS to determine the main range of the oligosaccharides of H.erinaceus.It came to the result that the extraction that is precipitated by 85%above of alcohol had the abundant of oligosaccharides.Then the H.erinaceus was extracted with boiling water and the method of alcoholic precipitation was used to remove the polysaccharides and get the crude oligosaccharides.It had a relatively high yield of 2.75 percent.The anion-exchange chromatography and the gel chromatography were performed on one after another to purify the crude oligosaccharides and we got 4 constituents.They were the salty constituent of HEOF-S-A and the aqueous constituents of HEOF-W-A, HEOF-W-B and HEOF-W-C.To determine the content and the structure of each constituent,the multiple techniques of HPLC,ESI-MS and GC-MS was used.And it's sure for the result that HEOF-W-A is a component with a molecular mass of more than 5000 Da and it's slightly soluble in water.HEOF-W-B is an oligosaccharide consisted of three arabinose with a molecular mass of 414 Da and it's a white crystal and is easily dissolved by water.And HEOF-W-C is a mixture of arabinose and mannose with a content ratio of 5.49 to 1.
