The codling moth, Cydia pomonella(L.), is one of the distinctly important quarantinepests of orchards, distributing all over the world. The larva of codling moth jeopardizes apple,pear, peach, walnut and so on, and cause the quality declining of fruit and financial loss offruit farmers. In the1950s, codling moth was found in Xinjiang province,China. Over the next40years, this pest spreads from Korla to the whole Xinjiang. In the middle of the1980s, withthe rapid development of tourism and transportation, codling moth had spread to Gansuprovince. Recent years, codling moth has been detected in Heilongjiang, Inner Mongolia andNingxia. and there is a great risk of codling moth proliferating. In this study, we carry on asurveillance in Gansu and Shannxi using pheromone-baited trap and field survey, to find outthe epidemic situation and occurrence tendency of codling moth. And we got the3’-terminalof Cydia pomonella odrant receptor2(CpomOR2) by reverse-translation polymerase chainreaction, and cloned this genetic fragment into pET-32a vector.The main results of this paper as follows:1.Field survey1:The field survey having occurred in Gansu province from May to September,2012,shows codling moth have not occurred in Tianshui city and1have occurred in Lanzhou city,while occurring in Wuwei, Zhangye relatively more than other areas.Although only1codling moth was caught in Lanzhou, we should strengthen themonitoring at all time,and then grasp the distribution of codling moth in Lanzhou, therebystrengthening surveillance to prevent invading from other areas.2.Field survey2:After three months of monitoring of the codling moth in Changwu and Binxian, and thenational highway G70, we have not caught the codling moth in anywhere, so Shannxiprovince is safe from July to September.Shannxi Province is adjacent with Gansu Province, and climate is similar with thelocality of codling moth. Along with the frequent transportation of agricultural products,codling moth will accelerated spread speed, so we should strengthen monitoring in Shannxiprovince, and thereby prevent codling moth invading. 3.Construction of prokaryotic expression vector:The3’-terminal of Cydia pomonella odrant receptor2was obtained byreverse-translation polymerase chain reaction with the cDNA of the antennae of codling moth,and this genetic fragment was cloned into pET-32a vector and integrated into E. coliBL21(DE3).
