Seabuckthorn(Hippophaerhamnoid.es), has two kinds of breeding methods,sexual reproduction and asexual reproduction. Asexual reproduction can maintain a single plant of high caste and bear fruit in advance2years,.in addition it can improve the propagation coefficient of more than20times, so it was used for the production nowadays. Asexual reproduction is divided into3types:hardwood, softwood and ramets tiller seedling propagation, tiller seedling of ramets was little used.But the disadvantages of hardwood and softwood was that a superior and clonal seedlings of selected was grown about20cm long segment once a year, then make cuttings limited3years later, so the propagation coefficient is low, greatly influenced the rapid propagation of seed, variety regional trial and establishment of the fertility orchard. So the methods and speed of hardwood and softwood was serious limited the propagation of seabuckthorn and the development of seabuckthorn industry. So, new asexual micropropagation method of seabuckthorn was needed to research. In this study, the Sea buckthorn branches was cut into5segments sequentially from the Top down, each segment is3cm.The segments were treated by five kinds of rooting hormone(ABTl,ABT2,GGR6,GGR7,and sodium nitroprusside+ABT1), respectively. In this study, three breeding methods (soil culture, hydroponics, sand culture) were adapted for growing the plant. And the physiological mechnaismsof rooting were explored. The results show that:rooting rates of the soil culture, hydroponics, sand culture were50%,65%,80%, respectively. The hightest rooting rates were the segments which were treated by GGR6and the rates in each matrice (soil culture, hydroponics, sand culture) were:55%,83.5%and96.8%. The third segment got the highest rooting rate of all five segments and the rates in each matrice (soil culture, hydroponics, sand culture) were:67.36%,73.75%,84.33%. At the same period, the average lengths of the Sea buckthorn roots that growed from three different matrices (soil culture, hydroponics, sand culture) were:1.9cm,3.6cm,6.45cm, respectively; the average root number were:2.1,2.75,4.45; and the average diameter of the roots were:2.35mm,2.6mm,3.25mm. Through observing the position of adventitious roots occur of seabuckthorn axillary bud in the process of cutting area, found adventitious roots of parts can be in leaf, stem, root and callus. Adventitious roots are mostly formed by the root primordial of roots, stems, and cambium of the main vein of leaves and then root primordial was splitted into adventitious roots and adventitious root location so, seabuckthorn type belongs to the induction of adventitious roots. Root primordium in the callus can be directly through the callus differentiation to form adventitious roots.Through detecting the Through detecting the14indicators by using14seabuckthorn plant hormones Ellisa kit,found that the content of soluble sugar was increased in the process of cutting, and the content of total nitrogen was cut down, C/N ratio was increased, the content of Arg was increased.The endogenous hormones of the bigger the ratio of IAA/ABA, the more beneficial to take root; the adventitious roots were promoted by the increased concentration of GA and CTK and reduced concentration of ETH; Rooting also was promoted by ETH.Enzyme system include SOD, POD, NOS, IAAO were related to the cuttings to take root. The content of SOD and POD was lower than before cutting in the process of rooting; The content of NOS was increased;The content of IAAO was elevated after cutting.In short, under the induced of rooting powder, in appropriate conditions of temperature,light,water,wet,and management, seabuckthorn axillary bud seedling root sprout is an easy, effective new technology. The success of this experiment, provided the theory and technical route based on operation for the industrialization and scale factory of seabuckthorn seedling.
