Cytoplasmic male sterility (CMS) is a main way to use rapeseed heterosis. The most promising CMS system is ogura CMS at present, The sterility of the system is very stable, and safe for hybrid seeds production, but the restorer gene exists in the radish only, it will carry a large segment of chromosome fragments from radish while transferring, which will lead to many bad agronomic traits of the generation. In1990s, by using the method of protoplast fusion, the scientists from INRA bred a new Ogura CMS successfully in France, which was called Ogu-INRA CMS. They also developed a several restorers with differential chromosome fragments from radish, for example R2000, the radish fragment is shortened, and can be used for commercial production. Unfortunately, this material has been patented. So to develop a new restore material is a major task for the application of Ogu-INRA CMS..The main materials in this study were including CLR650,SR,YR,CR (from Hunan),7-2140,7-2143,7-7302,7-7304,7-7311,7-7314(from Professor Tu) et al. In order to identify whether CLR650was new Ogura CMS restorer, molecular biology research tools were used in this study, including analysis of specific markers and linkage group analysis. To observe the transfer of the restore gene in CLR650, The method of cytology was used. The main conclusions were as following:(1)The restorer gene in CLR650located in the linkage group N19, and similar to the other restorers. However, the markers around the restore gene in the material CLR650were different compared to above restore sources. The pathwayto introduce the restore gene inabove sources was obtained by protoplast fusion, and CLR650was transferred by sexual hybridization, and the donor material of restorer gene Rfo was also different.(2)Through the analysis of molecular markers linkage with Rfo on chromosome in radish, the radish chromosome fragment length in CLR650was significantly shorter than similar foreign restorer.(3)The chromosome number of CLR650was basically38.(4)The segregation ratio in the offspring generated by CLR650combinations did not match with the rules of the Mendelian. We speculated the phenomenon that was caused by chromosomes bridges during meiosis. In the experiment of pollen germination, the germination of pollen of CLR650are basically the same with the contrasts, so the viability of male gametes of CLR650is normal.
