Lycoris is one of the important ornamental flowers. Research on Lycoris crossbreeding and culture of the filial generation has the vital significance on interspecific relationship, origin and new variety breeding of hybrid.L.sprengeri L.longituba, L.chinesis, L.radiata, L. haywardii, respectively, were selected for hybridization. Examination of pollen vitality, cross experiments, culture of the filial generation and seed propagation of Hybrids were conducted. The results were as follows:1. Admeasure the pollen viability of Lycoris by acetate-carmine staining (92.55%) TTC staining (25.45%), I2-KI staining (77.11%) and germination method (73.96%) Significant difference exists among four methods through the analysis of variance. I2-KI staining get close to germination method on the value. It could be a simple method for admeasure the pollen viability of Lycoris.Pollen germination rate gradually decreased with the extension of storage time, the trend of reduce on the basis of different species. The pollen vitality of5kinds of pollen declined significantly, below50%, after stored6days. Around15d, L. radiata pollen fell below10%, others remained at20%~30%after21d, all of them below10%after srored50d. The different storage methods had significant effects on the pollen germination of different species; the best storage method is4℃dry storage. The pollen vitality of L. haywardii is40.95%after15d. The strength of pollen vitality of5kinds of pollens is:L. haywardii, L.sprengeri, L.longituba, L.chinesis, L.radiata.2. The rate of seed setting and the rate of inflated germen vary with the different pollination combination of Lycoris. The rate of inflated germen of L.sprengeri, L.chinesis, L.longituba, and L.haywardii was between72%and90%, and the Pollination combination of L.haywardii and L.longituba was can reach to90%. Seed setting rate appeared very different, in addition to by the L.haywardii as female parent hybrid with L.longituba and L.sprengeri which seed setting rate were78%and85%respectively, the rest of the combination of seed setting rate was low between20%and38%. Lycoris radiata hybrid, respectively, with L.longituba, L.sprengeri, L.haywardii, L.chinesis of ovary enlargement, the rate of Inflated germen was low between23%and42%, seed setting rate was low between3%and7%.The days from pollination to inflated germen fade varies with the different pollination combination of Lycoris between18and30d. Female parent plays a decisive role at the rate of seed setting and the rate of inflated germen, in proper order was:L.haywardii, L.sprengeri, L.chinesis, L.longituba, and L.radiata. Different cross combinations using different pollination methods, had a significant effect on the Rate of Inflated Germen, L.radiata♀x L.longituba♂, L.radiata♀x L.sprengeri♂were suit with conventional method or the cutting style pollination method, L.radiata♀x L.chinesis♂was more suitable for using pollen mixture method, L.radiata♀x L.haywardii♂was more suitable by conventional pollination method.3. Disinfection mode of inflated germen the pollination combination of Lycoris radiata and L.sprengeri was75%ethanol to wipe3times+75%mercuric chloride soak10min+2%sodium hypochlorite soak20min. the optimum medium formula was2,4-D2.0mg/L+NAA1.0mg/L+6-BA0.5mg/L.Ovary slice culture can be performed after hybrid pollinated5d、10d、20d、30d、40d. The survival rate increases with time increasing, pollination30-40d was to achieve the best effect. The survival rate of ovary slice culture of the cross combination of L.radiata and L.haywardii could reach65%. Ovule culture could be performed after hybrid pollinated5d、10d、20d、30d、40d、50d. The survival rate of ovule culture into a clear upward trend with the time increasing.The survival rate of ovule culture of the cross combination of L.haywardii and L.sprengeri can reach90%. Embryo culture could be performed after hybrid pollinated30~40d、40-50d、50-60d. The success rate of embryo culture which along with the growth of the hybridization combination pollination continuously rises,40to50days reach the highest. The survival rate of embryo culture of the cross combination of L.longituba and L.sprengeri can reach55%.4. The seed germination rate of L. haywardii, L.sprengeri and L.longituba are all over90%. The seed germination rate of hybrid combination of L.haywardii and L.sprengeri, L. haywardii and L.longituba, L.sprengeri and L.longituba, L.longituba and L.sprengeri are between80%and90%. The time of hybrid leaf consistent with female parent. The hybrids of L. haywardii and L.sprengeri, L. haywardii and L.longituba as same as L. haywardii are belong to the autumn leaf. Leaf time concentrated in early October. The hybrids of L.sprengeri and L.longituba, L.longituba and L.sprengeri as same as L.sprengeri, L.longituba were belong to the spring leaf. Leaf time concentrated in early February. Blade width unchanged since begin to leaf, always keep one piece. But leaves present regular growth trend, the growth of hybrids and their parents were consistently could be divided into initial stage, growing stage, after part growth stage. The elongation of leaf was50%-60%of the total leaf high at growing stage.
