Microsatellite DNA was often used in wildlife forensics for identity recognition and paternity test. Standardization of genotyping is the key for the application of microsatellite. Many researches on microsatellite analysis of tiger (Panthera tigris) have been reported, but non of the genotyping was standardized.8tetranucleotide microsatellite locus of domestic cat (Felis catus), which are F85, FCA441, F124, F53, C09, C12, D06and B04, were proved to be feasible on tiger by our former researches. The structure of the8loci were sequenced and analyzed, and then named follow the STR genotyping rules of ISFG committee. Results showed that the core structure of C12and B04are complex, and different alleles showed the same genotype. Therefore, it suggested that locus C12and B04be eliminated from the paternity test system, and the rest loci are completely usable. In addition, the8loci were used to analyze a Amur tiger population. Number of alleles (Na), Allele frequency, number of effective alleles (Ne), observed heterozygosity (Ho) and expected heterozygosity (He) were obtained, all loci were tested for Hardy-Weinberg equilibrium, multilocus heterozyosity (MLH) and standard individual heterozygosity (SH) were calculated, inbreeding coefficient and inbied individual correlation analysis of the tiger population were calculated by COANCESTRY. Results showed that genetic diversity of the population is relatively high, and there is mechanism to avoid inbreeding. But the inbreeding was not completely stopped; there is a still risk for loss of genetic diversity. Effective strategy for population genetic management was needed.
