To reveal the genetic characteristics of cashmere economic traits,756cashmeregoats in China, four cashmere goat groups (Hexi cashmere goat, Inner Mongoliacashmere goat, Liaoning cashmere goat, Huanxian cashmere goat) were used asexperimental populations. KAP13.1and KAP13.3gene was amplified usingPCR-single stranded conformational polymorphism (PCR-SSCP) analysis to detectgenetic variation. At the same time using Least square analyzed its associations oncashmere yield(kg), body weigh(tkg), cashmere thicknes(smm), cashmere fineness(μm)traits of Liaoning cashmere goats, The result as follows:(1)The PCRamplification obtained KAP13.1gene with491bp. Five nucleotide polymorphismsites (A-D), which mainly came from point mutation, were identified in four alleles.There were three transition sites (60%) and two transversion sites (40%). Themutation of170and197'TG, C'G resulted in Leu'Arg, Thr'Ser missensemutation. Four alleles were found in Hexi and Huanxian cashmere goat populations,while there were no allele C and D in Inner Mongolia cashmere goats, and allele Dhad not been detected in Liaoning cashmere goat populations. Clustering resultsshowed that Hexi and Huanxian cashmere goat populations clustered together,Liaoning and Inner Mongolia cashmere goat populations clustered together. Thepolymorphic locus of KAP13.1gene significantly affect on cashmere yield and bodyweight in Liaoning cashmere goat, and genotype AB cashmere yield and body weightwas significantly more than genotype AA.(2)The caprine KAP13.3gene wasamplified with559bp. Sequencing of the amplicons confirmed the presence of eightdifferent sequences (A-H), all of these showed a high sequence homology to knownovine KAP13.3, bovine KAP13.3and caprine KAP13.1sequences. Nine nucleotidesubstitutions were identified within the KAP13.3gene coding region, and all of thesewere non-synonymous. All sequences showed an open reading frame of471nucleotides encoding156amino acids. This polypeptide contained high levels ofserine (23.08–23.72mol%), cysteine (10.26–11.54mol%), and threonine (5.66mol%).The polymorphic locus of KAP13.1gene significantly affect on cashmere thicknessand body weight in Liaoning cashmere goat, and genotype EH cashmere thicknesswas significantly more than genotype AA, genotype AE body weight was significantly more than genotype AA, EH, DE. The results showed that all of the four cashmeregoat populations are moderately polymorphic in KAP13.1gene, and there are richgenetic diversities among the four cashmere goat populations. KAP13.1gene is thecandidate gene with affecting cashmere yield and body weight trait of Liaoningcashmere goat. AB genotype can be the marker genotype of multi-trait (cashmereyield, body weight) marker in assistance selection. The caprine KAP13.3gene ispolymorphic and it is the candidate gene with affecting cashmere thickness and bodyweight trait of Liaoning cashmere goat. EH can be the preferred marker genotype ofcashmere thickness, AE can be the preferred marker genotype of body weight inassistance selection.
