The microbiology laboratory of Soil and Fertilizer Research Institute of Sichuan Academy of Agricultural Sciences successfully cultivated new strain258with parents spawn53and spawn West33by using monosporous hybridization technology since2009to2011and it could replace the major production variety of Sichuan Province. To authenticate strain258and research its biological characteristics is very important to the development of Pleurotus cornucopiae industry in Sichuan Province.We used the method in the national standard《Identification of distinctness for edible mushroom cultivar by antagonism》 and 《Verification of genuiness for edible mushroom spawn-RAPD》 to authenticate strain258from spawn51、spawn53、spawn West33、 Sichuan Pleurotus cornucopiae2, according to the national standard 《Registration of crop variety edible mushroom》and did Production test and Regional test of strain258comparing with spawn West33to Statistics its High yield and Yield stability indicators. Then we also did biological characteristics test of strain258to find its optimum growth conditions and difference from the control strains.The results following:The antagonistic reaction results showed obvious antagonism between the5strains, and the5strains were not antagonistic reaction itself. It can be concluded that the5strains are different varieties.We screened out6RAPD marker primers that which have clear and diversity amplified bands, and amplified41clear and polymorpHic bands.After that used the NTSYSpc2.1software to analysis Phylogenetic tree and Genetic distance.The result showed that the genetic similarity coefficient of the five strains is between0.5488to0.7805;when the similarity coefficient is at0.7226,5strains were clustered into4groups; each for a class of spawn West33、strain258、Sichuan Pleurotus cornucopiae2、spawn51and spawn53clustered together, the genetic distance of strains258with the control strain Pleurotus cornucopiae spawn51、spawn53、spawn west33、Sichuan Pleurotus cornucopiae2is respectively0.2428、0.2744、0.4469、0.3311; It showed that strain258is a new variety of Pleurotus cornucopiae and is different with control strains.The production test and regional test results showed that strain258had good stability、high yield in different areas and in Mass production. The strain258’s average yield compared to the control spawn was above20%, the yield difference reached significant level.The strain258could completely replace the spawn West33and have high popularization value.The results showed that strain258is as the same as control strains in most biological characteristics that because they belong to the same species. The strain258has significant differences to control spawn at the optimum pH (pH6) and the optimum temperature (30℃) as well as the carbon source. Biological characteristics of strain258was also proved the optimum growth conditions were:pH6, temperature30℃, soluble starch as carbon source, peptone as the nitrogen source...
