The Radix Isatidia Polysaccharides (RIP), extracted from the traditional Chinesemedicine Radix Isatidis, were orally administrated to weaning Sprague-Dawley rats indifferent group. The rats were randomly allocated into the following groups, control(group A), mid dose RIP (group B), high dose RIP (group C). The technologies,including anatomic, morphological, immunohistochemistry and serum absorbingexperiments, were employed to evaluate the assimilation, morphology anddevelopment of mall intestine, the number of intestinal epidermal growth factor(EGF)-positive and growth hormone release inhibitory hormone (SS)-positive cells inrats. Rats aged28,35,42,49days were selected to assess the intestinal developmenteffect of RIP on weaning rat, so as to clarify the potential mechanism and exploithealthy RIP. The results were listed as follow:(1) RIP was non-nutritional polysaccharides. RIP administration had nosignificant on the body weight, small intsestine weight of weaning rats, but couldincrease the length of small intsestine significanyly at35days (P<0.05).(2) RIP could increase the villus height, decrease the crypt depth, and increase thevillus height to crypt depth ratio(V/C ratio), thus improving the development of smallintestine in weaning rats. The optimal dose that could affect the villus height, cryptdepth, and V/C ratio increased with the age of the rats.(3) RIP could improve the D-xylose absorptivity of weaning rats significantly.(4) RIP could improve the secretion of mucosal EGF-positive cells and increasethe mucosal EGF-positive cells in duodenum of weaning rats.(5) On the contrary, RIP could suppress the SS secretion in duodenum andjejunum of weaning rats by reducing the number of SS-positive cells. In conclusion, as non-nutritional polysaccharides, although RIP administrationhad no significant on the body and small intestine weight, the other improvements ofRIP on weaning rat intestinal health, including increasing the weight of smallintsestine, enhancing the assimilation, improving the development of small intestine,promoting the secretion of mucosal EGF-positive cells, suppressing the SS secretion,have been observed in the research.
