In this study, Wannan Black pigs were used to detecting po lymorphism of SCAP geneand CETP gene-candidate genes associated with blood cholesterol levels-by PCR-SSCPmethod, then analyzed the association with gene polymorphisms and blood cholesterollevels by the SPSS program GLM model. In addition, RT-PCR technique was detected tostudy the difference of ABCA1gene expressions in the liver, heart and other different tissues.The aims o to provide certain basis for protection and exploitation in Wannan Black pigsand provide the theoretical basis for fostering low-cholesterol pigs breeding.The results show that:(1) a G→T mutation (G813T) occurred in813base position of exon2of SCAP inWannan Black pigs, one mutational genotype (TT) was detected in G813T site, and thismutation caused amino acid change from methionine to isoleucine. The TT genotypefrequencies is1.0, T allele gene frequency is1.0.(2) a C→T mutation (C795T) occurred in795base position of intron1of CETP inWannan Black pigs,3genotypes (CC, CT and TT) were found in C795T site. Genotypicfrequency of CC, CT and TT was0.6250,0.3036and0.0714respectively, gene frequencyof C and T was0.7768and0.2232respectively. the polymorphism information content(PIC) was0.2866in Wannan Black population, which indicated it was moderatepolymorphism. The χ2value was1.7388, it was in Hardy-Weinberg equilibrium in WannanBlack population (P>0.05).(3) the statistical analysis found that the relationship between HDL-C and TC levelsand different genotype individuals in C795T site existed the significant difference(P<0.05), while the relationship between LDL-C and TG levels and different genotypeindividuals in C795T site existed the non-significant difference.(4) ABCA1gene in different tissues of Wannan black pigs had different levels of geneexpression, the highest expressionwas in the liver, followed was in the lung, and kidney wasthe lowest level of gene expression; the levels of gene expression in the liver and lung werevery significantly higher than them in other tissues present(P<0.01), and the levels ofgene expression in the heart were significantly higher than them in the kidney and spleen(P<0.05).(5) ABCA1mRNA expression in the liver was significantly negatively correlated withTC levels of Wannan black pig, the correlation coefficient was-0.913; ABCA1mRNAexpression in the liver was moderately low negatively correlated with LDL-C levels, the correlation coefficient was-0.475.
