Study Of Asexual Propagation Techniques Of Styrax Tonkinensis

Styrax tonkinensis is a deciduous tree from Styracaceae with the value of medicine、oil、timber andornamental. In this paper, obtaining aseptic plant by means of aseptic seeding, using parts of the plantstems as explant to study the effect of vitro culture of Styrax tonkinensis. The different plant hormonesin the stage of proliferation culture, as a result, the best mediums of every stage were screened out,tissue culture system for Styrax tonkinensis was established. Studying the survival rate come fromdifferent stocks、grafting time、grafting methods and stock age in the grafting experimnent. In addition,the landscape application of Styrax tonkinensis was preliminary discussed. In this study, it can provide atheoretical and experimental basis for the rapdi propagation of Styrax tonkinensis.The main results as following：1．Establishment of rapid propagation system.The results showed that the disinfection method of seeds which75％alcohol disinfected20secondsfirstly, secondly deeped in5.0g·L^-1NaCLO to disinfect15minutes, thirdly washed for5times withsterile, lastly cut the endospeerm of seeds inoculated into MS medium, can make the survival rate90%and contamination rate5%. The best proliferation of the shoots was5.7times, and the average height ofshoots was4.46cm, when adding0.3mg·L^-1BA to MS medium, the shoots grew much and strong.Twostep rooting method,to treat the shoots with100mg·L^-1IBA for3days, and then transfer them into1/2MS medium could otain60%of rooting rate. The best time of hardening plantlets was8-12d,transplanted in the artificial mixture with the ratio of perlite:vermiculite:peat （1:1:1）, and survival ratewas the highest.2. Study of grafting propagation.The effect of different rootstocks、grafting time、grafting methods and stock age on Styrax tonkinensis ’ssurvival ratio were studied. It indicated that, taking the scions of wax and cold treatment grafted onannual and vitality stocks in the late April is the optimal combation of grafting factors, survival rate isabout78%. Scion wiht buds、management method and grafting method is not the key factors,Graftingtime、scion treatment and stock ageis the main influence factors of the grafting survival rate.3. Landscape application discussion. Styrax tonkinensis is an excellent ornamental plant with white flowers、florescence4-6month andfrangrance. It can be used as a graden ornamental plant、shade tree、landscape forest and one specialtype of graden of Styracaceae species.
