Study On Application Of Chinese Herbal Compound Extraction In Subsituting Feed Antlbiotics | | Posted on:2014-12-22 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Q Yang | Full Text:PDF | | GTID:2253330425952870 | Subject:Food engineering | | Abstract/Summary: | PDF Full Text Request | | This study was designed to investigate the extract of compound Chinese medicinalherb(TSKZâ… and TSKZâ…¡) that was developed in our group which had effects onpathogenic bacteria and probiotics susceptibility by the method of bacteriostatic test invitro, and compared with feed antlbiotics;then through the breeding test to study theeffects of TSKZâ… and TSKZâ…¡on broiler performance, meat quality, immune functionand intestinal flora in broilers,in order to study the two compound Chinese herbalextracts whether exit the possibility of alternative feed antibiotics on broilers.We conducted bacteriostatic tests in vitro by Oxford-cup method on Escherichiacoli,Salmonella enteritidis, Salmonella aertrycke, and Staphylococcus aureus to pick outbroad spectrum bacteriostatic herbal medicines, while diameter of inhibition zone wasused as the evaluating indicator; on the other hand, Hungate anaerobic technique wasemployed to single out herbal medicines which do not affect growth of2probiotics(Lactobacillus acidophilus and Bifidobacterium), while colony forming units(cfu) was taken as estimating parameters.The breeding test selection140one-day-old Rose308broliers were randomly assignedinto7groups with2replications after7days feeding. the water was supplemented with5mL/Lã€10mL/Lã€20mL/L of TSKZ â… a nd5mL/Lã€20mL/L of TSKZâ…¡ to feedexperimental animals,blank and feed antibiotics (chlortetracycline,50ppm)for controlgroups. The experiment perid were42days. The resultswere as follows:1. The bacteriostatic test in vitro experiments showed that, the extract of compoundChinese medicinal herb inhibited the growth of pathogenic bacteria and promoted thegrowth of probiotics simultaneously, effects of TSKZâ…¡ were the best among them.Butfeed antlbiotics inhibited both pathogenic bacteria and probiotics.2. The Chinese herbal medicine could promot the growth and improved the meatquality(1) Under the experimental conditions, the better average daily gain was gotten whenthe addition of Chinese herbal medicines(P<0.05), compared with the control group ofchlortetracycline were no significantly difference(P>0.05), Which TSKZ II high dosegroup increased of22.12%that was the most obvious effect; the low-dose group TSKZ II’s average daily feed intake is lower than the control group; the low-dose group ofTSKZ I and low-dose,high-dose of TSKZ II compared with the control group reduced9.60%,12.50%,9.10%(P<0.05),compared with the control group of chlortetracyclinewere no significantly difference(P>0.05);Chinese herbal compound extracts increasedthe survival rate of broilers.(2) When the TSKZâ…¡additive was high-dose,the hardness of chicken breast musclesignificantly increased(P<0.05);when TSKZâ… additive was high-dose,the elasticity andchewiness of chicken breast muscle significantly increased compared with the controlgroup(P<0.05);the group of high-dose of TSKZâ… and the chlortetracycline group couldincreased the brightness of chicken breast muscle(P<0.05).3. Effects of herbal medicament extracts on broilers of immune function(1) Effects of compound Chinese medicinal herb on immune organ index in broilers.The TSKZâ…¡in which the relative quality of thymus significantly increased comparedwith the control group(P<0.05),The low-dose of TSKZâ…¡increased the relative qualityof spleen(P<0.05) at the age of21d;the high-dose of TSKZâ… and TSKZâ…¡significantlyincreased the relative quality of thymus,the middle and high dose of TSKZâ… increasedthe relative quality of bursal.(2) Effects of compound Chinese medicinal herb on NewCastlediseas in broilers. Inthe22~42d the best antibody titer of Newcastle disease in broilers was increased by24.40%when the addition of a high level of TSKZâ… (P<0.05), antibody level wasenhanced to some extent by Chinese medicinal herb. From the whole test results,therewas no significantly difference between the groups of Chinese medicinal herb andchlortetracycline(P>0.05).(3)Effects of compound Chinese medicinal herb on splenic lymphocyte proliferation inbroilers.the high dose of TSKZâ… andâ…¡significantly increased compared with the controland chlortetracycline(P<0.05) at the age of21d; the middle and high dose of TSKZâ… and TSKZâ…¡ raised obviously splenic lymphocyte proliferation by30.43%,43.48%,39.13%,26.09%compared with the control and chlortetracycline(P<0.05). In general,the effect of high-dose compound herbalâ… was better than other groups.4. Effects of compound Chinese medicinal herb on intestinal flora in broilersThe results indicated that the number of E.coli in the administrated groups andchlortetracycline were significantly inhibited(P<0.01) that compared with the controlgroup, the number of lactic acid bacteria were significantly increased by high-dose ofTSKZâ… and low-dose of TSKZâ…¡ (P<0.05),It is obvious that chlortetracycline showedan decreasing trend(P<0.01)that compared with the control group.Conclusion:Compound Chinese medicinal herb could enhance the productionperformance, compared with the control group of chlortetracycline were no significantlydifference(P>0.05); in improving meat quality, enhance the immune function of broilers and improve broiler gut microbiome were better than chlortetracycline.
