Study On Expression Change Of SGLT1and GLUT2mRNA In Intestine Of Hy-line Hens At Growing Stage And The Relation With Production Performance

The experiment aimed to investigate expression developmental pattern ofsodium/glucose transporter1(SGLT1) and glucose transporter2(GLUT2) mRNA and therelationship between expression developmental pattern and production performancethrough studying expression of SGLT1and GLUT2mRNA in growing Hy-line Hens’intestine and determining the data of production performance at the same time. Onehundred and fifty57-day-old health and same genetic background Hy-line Hens wereselected and had been fed for10weeks. Six hens were picked up from each fixed growthperiod respectively such as57days,71days,85days,99days,113days and127days.Those hens were killed and small intestine mucosa were separated for samples which weredetermined by fluorescence quantitative PCR. What’s more, production performance wererecorded at every weekend. The result are as follows:1. Relative expression quantity of SGLT1mRNA in Hy-line Hens’ intestine mucosa atthe stage of growing was3.26×10-5at57d, reduced at71d compared with57d, increasedfrom71d to99d, reduced from99d to127d and the maximum was10.37×10-5at99d. Therelative expression quantity of SGLT1mRNA at71d reduced59.51%compared with57d(P<0.05), increased75%at85d compared with71d(P<0.05), increased348.92%at99dcompared with85d(P<0.05), reduced59.50%at113d compared with99d(P<0.05), reduced21.67%at127d compared with113d(P<0.05) and was not significant with that at57d(P>0.05).2. Relative expression quantity of GLUT2mRNA in Hy-line Hens’ intestine mucosaat the stage of growing was2.79×10-5at57d, reduced from57d to85d, increased at99dcompared with85d, reduced from99d to127d and the maximum was9.80×10-5at99d.The relative expression quantity of GLUT2mRNA at71d reduced19.71%compared with57d(P<0.05),reduced42.41%at85d compared with71d(P<0.05), increased659.69%at99d compared with85d(P<0.05), reduced59.90%at113d compared with99d(P<0.05),reduced38.93%at127d compared with113d(P<0.05) and was not significant with that at71d(P>0.05).3. From57d to71d, relative expression quantity of SGLT1and GLUT2mRNA bothreduced significantly(P<0.05), average daily feed intake(ADFI), average daily gain(ADG)and feed to gain all increased and were not significant(P>0.05); from71d to85d, relative expression quantity of SGLT1mRNA increased significantly(P<0.05), relative expressionquantity of GLUT2mRNA reduced significantly(P<0.05), ADFI, ADG and feed to gain allincreased and were not significant(P>0.05); from85d to99d, relative expression quantityof SGLT1and GLUT2mRNA both increased significantly(P<0.05), ADFI and feed to gainincreased significantly(P>0.05) and ADG reduced significantly(P>0.05); from99d to127d,relative expression quantity of SGLT1and GLUT2mRNA both reducedsignificantly(P<0.05), ADFI increased with no significant difference(P>0.05), feed to gainincreased significantly(P>0.05) and ADG reduced significantly(P>0.05).The result showed that there were different developmental pattern between SGLT1and GLUT2mRNA in growing Hy-line Hens’ intestine, but they both had relation toproduction performance. From57d to85d, ontogenetic expression of SGLT1and GLUT2mRNA were in line with representation of production performance; from85d to99d,ontogenetic expression of SGLT1and GLUT2mRNA were in line with ADFI and feed togain and were different with ADG; from99d to127d, ontogenetic expression of SGLT1and GLUT2mRNA were in line with ADG and were different with ADFI and feed to gain.
