This study is mainly investigated the preservation effect of cock semen diluents at4℃in five kinds of diluents, namely sodium glutamate、L-carnitine、betaine、D-sorbitol and trehalose added into the buffered of phosphate, respectively. The preservation effect is evaluated by five evaluation indicators of semen live rate, survival time, survival index, deformity rate and rate of acrosome intergrity in order to choose the optimum concentration of each dilute. The detailed contents and results are as follows:For the addition of sodium glutamate experimental group, the results show that the best concentration of sodium glutamate is1.6g/100mL. The evaluation indicators are shown that semen live rate is0.5at42h and survival index is31.55at104.5h; both of which are higher than the group of2g/100mL (P<0.05), significantly higher than1.2g/100mL and0.8g/100mL(P<0.01).For the addition of L-carnitine-1.6g/100mL sodium glutamate experimental group, the results show that the best concentration of L-carnitine is7.5mg/100mL. The evaluation indicators are shown that semen live rate is0.5at42h and survival index is34.35at126h; semen live rate and survival index are significantly higher than others(P<0.01).The effect of betaine-7.5mg/100mL L-carnitine-1.6g/100mL sodium glutamate on semen vitality has been studied through experiment. The results show that the best concentration of betaine is100mg/100mL. The evaluation indicators are shown that semen live rate is0.5at115h and survival index is86.11at177h which is higher than the group of80mg/100mL(P<0.05), significantly higher than40mg/100mL and60mg/100mL(P<0.01).For the different concentration D-sorbitol-7.5mg/100mL L-carnitine-1.6g/100mL sodium glutamate diluents, the results show that the best concentration of D-sorbitol is20mg/100mL. The evaluation indicators are shown that semen live rate is higher than0.5at48h and survival index is43.20at126h; survival time is higher than others(P<0.05).The effect of trehalose-7.5mg/100mL L-carnitine-1.6g/100mL sodium glutamate on semen vitality has been studied. The results show that the best concentration of trehalose is100mg/100mL. The evaluation indicators are shown that semen live rate is0.6at73h which is higher than the group of80mg/100mL(P<0.05), significantly higher than40mg/100mL and60mg/100mL(P<0.01). The survival index is61.80at126h which is higher than the group of80mg/100mL(P<0.05), significantly higher than40mg/100mL and60mg/100mL(P<0.01).In the whole, nineteen kinds of diluents have no obviously influence on the deformity rate and rate of acrosome intergrity. For various additives, the preservation effect of betaine is the best in five kinds of additive. In addition, the optimum concentration is100mg/100mL betaine-7.5mg/100mL L-carnitine-1.6g/100mL sodium glutamate for diluents.
