Microsatellite instability (MSI) has been used widely to detect the genome of tumor of human. Reticuloendotheliosis (RE) is the third chicken tumor disease after the Marek’s Disease and the Avian Leukosis. It has not been reported that detecting genomes changes of chicken embryos cells infected with reticuloendotheliosis virus (REV) by microsatellite technology. It aims to detect whether the cells’ genome changes and the genome alteration at the early stage of the chicken embryos fibroblasts infected with Reticuloendotheliosis Virus by the microsatellite markers on the chicken chromosomes.In this study,22groups of chicken embryo fibroblasts (CEF) were prepared from22SPF chicken embryos. In each group, one was infected with REV, the other one was no-infected as control. DNA was extracted from the CEF. All DNA were amplified with the polymerase chain reaction by the135microsatellite markers on the chromosome1,2and3. All products were tested by12%denatural gel electrophoresis and silver stain to detect the MSI of the genome.106microsatellite markers which showed different percentages of MSI accounted for78.52%of the135chosen microsatellite markers. The highest ratio of MSI was50.00%. There are16microsatellite loci whose ratio of MSI is more than30%.29loci did not occur MSI.Microsatellite instability (MSI) was found in all of22groups. The highest ratio of MSI was the group3, which is40.00%. The ratio of MSI in the others was under30%. The other groups’MSI are under30%, including3groups (1,2,13) which ratio was between20-30%,13groups which ratio was between10-20%and5groups which ratio was under10%.The MSI was first found in chromosome1,2and3of CEF infected with REV. This thesis indicated the same result. The MSI in this article may be suggested as a valuable method for the etiopathogenesis and early diagnosis of the reticuloendotheliosis.
