Jasmine [Jasminum sambac(L.)Aiton], a evergreen undershrub belonging to Oleaceae family,originated India and Pakistan. In this study, the material is double-petal jasmine, With the colchicin to treat jasmine’s growing point, the induction effect by different densities and the treating times of dropping liquid method、smear method and soaking method had been studied. According to the chromosome number examination ploidy appraisal method and flow cytometric analysis, the tetraploid plants were obtained, morphological change rate、the budding rate and variation rate with colchicin were contrasted systematically. The study expected to find out a serious of suitable inductive methods of Jasmine polyploid and to make rich of the resources of Jasmine foundationally.The main research results were as follows:1The annual Jasmine cutting seedlings, planted in the greenhouse of Chongqing University of Arts and Sciences. A month later, when the jasmine survived, prun them to promote the multiple shoots, and fertilizate them once per week.2Study on polyploid induction and its identification by chemical induction of somatic cells of Lonicera japonica Thunb.3. Ploidy tests of induced plants in the initial stage can be carried out through observing appearance of plants. Compared diploid Jasmine with tetraploid Jasmine, the induced tetraploid plants showed that leaf index becomes larger, darker. At the same time the mutate plantlets were deformed and the internode lengthes were shorter than the average of normal Jasmine’s. Those characters of Jasmine could be taken as the advance index in testing tetraploid Jasmine. 4. The anatomy research shows, stoma and guard cells increase, stomata number in unit area decreased.5. According to the cytological study, the chromosome number in the polyploidy plants are2n=2x=26, but in the mutants are2n=4x=52. The relative DNA content of diploid was1500, mutants was3000by flow cytometry analysis to determine their ploidy.6. Physiological characteristics comparison results showed:the SOD content, POD content, proline content, soluble sugar content of Jasmine variant were higher than the diploid, while the MDA content was lower than diploid, and the differences were significant.
