Objective To explore the interleukin-1α and its receptor antagonist effect on theinvasion and proliferation in human colon cancer cells with different metastatic potentials aswell as the role of IL-1α and IL-1ra in metastatic process.Method RT-PCR was used to detect the expression of IL-1α and IL-1I type R mRNAin colon cancer cell lines with different liver metastatic potential. The secreted levels of IL-1αby colon cancer cells were measured by ELSIA.We also examined how IL-1α and IL-1rainfluence the proliferation and invasion in colon cancer cells with different liver metastaticpotential using the premix WST-1assay and Invasiveness assay,respectively.Result RT-PCR results showed that IL-1I type R were detected in all colon cancer cellline. IL-1α mRNA were detected in highly metastatic colon cancer cell line(85±0.520pg/ml).Recombinant IL-1α significantly enhanced growth and invasiveness of theall colon cancer cells (P<0.01). Moreover,IL-1ra could inhibit the proliferation andinvasiveness of colon cancer cell line significantly (P<0.01).Conclusion IL-1α may play a role in liver metastasis of colon cancer via enhancedcolon cells proliferation and invasion; therefor,IL-1ra can significantly inhibit the coloncancer in proliferation and invasion in WiDr cells. |
