Abstract/Summary:
Objective To investigate an association between heat stress protein70-1(HSP70-1)-110,+190and Tumor necrosis factor-α (TNF-α)-308gene polymorphism and metabolic syndromeand its related indicators among Hui and Han nationalities in Ningxia; To investigate therelationship of occupational psychological stress level and metabolic syndrome and withHSP70and TNF-α gene polymorphism.Method According to a1:1matched case-control study design, workers of regionalorganizations and institutions aged20-60who received medical health checkup and endocrinepatients in General Hospital of Ningxia Medical College and WuZhong People’s Hospitalfrom October2011to October2012were enrolled in the study. According to the inclusionand exclusion criteria,600patients were identified as a case group,150men (25%) and150women (25%) of Hui,150men (25%) and150women (25%) of Han, mean age (50.38±6.82years); Matched by age, sex and ethnic, population from the same period in General Hospitalof Ningxia Medical College and WuZhong People’s Hospital out-patient clinics for regularhealth check-randomly selected600healthy people in the control group, mean age(50.38±6.78years). Data were collected through questionnaire, physical examination andlaboratory examination, and Minor Groove Binder-Taqman (MGB-Taqman) probe methodwas applied to detect the single nucleotide polymorphisms of HSP70-110,+190and TNF-α-308.Results There were no significant differences in the distribution of genotypes andallele frequencies of HSP70-1+190between the metabolic syndrome group and control group(all P>0.05), there were significant differences in the distribution of genotypes andallele frequencies of HSP70-1-110and TNF-α-308between the metabolic syndrome groupand control group(all P<0.05); There were no significant differences in the distribution ofgenotypes and allele frequencies of HSP70-1-110,+190and TNF-α-308between Hui andHan nationalities(all P>0.05); There were no significant differences in the distribution ofgenotypes and allele frequencies of HSP70-1+190among abdominal obesity with metabolicsyndrome, non-abdominal obesity with metabolic syndrome group and control group(allP>0.05); There were significant differences in the distribution of allele frequencies ofHSP70-1-110among abdominal obesity with metabolic syndrome, non-abdominal obesitywith metabolic syndrome group and control group(P<0.05), compared with abdominal obesitywith metabolic syndrome group, control group had significant differences in the C alleledistribution of HSP70-1-110(χ~2=6.431,P=0.011,OR=1.141); There were significantdifferences in the distribution of genotypes and allele frequencies of TNF-α-308amongabdominal obesity with metabolic syndrome, non-abdominal obesity with metabolicsyndrome group and control group(all P<0.05), compared with abdominal obesity,non-abdominal obesity with metabolic syndrome group, control group had significantdifferences in the A allele distribution of TNF-α-308(χ~2=20.422,P=0.000,OR=2.051andχ~2=8.514,P=0.004,OR=1.913); There were significant differences in the distribution of thelevel of occupational stress between the metabolic syndrome group and control group(P<0.05),Adjusted for age, marital status, education level, smoking and drinking, the odds ratios forMS were1.866and2.536in the people with moderate and severe stress, respectively; Therewere no significant differences in genotype and allele frequencies of HSP70-1-110,+190andTNF-α-308in the three stress levels (all P>0.05); By comparing the clinical data of differentgenotypes, we find that there were no significant differences in WC, WHR, BMI, WBC, SBP,DBP, TG, TC, HDL, LDL, FPG and UA (all P>0.05), but there were significant differences inAST and ALT of the genotype distribution of HSP70-1-110(all P<0.05); There were no significant differences in WC, WHR, BMI, WBC, SBP, DBP, TG, TC, HDL, LDL, FPG,AST, ALT and UA of the genotype distribution of HSP70-1+190(all P>0.05); There were nosignificant differences in WBC, SBP, DBP, TG, TC, LDL, AST, ALT and UA (all P>0.05),but there were significant differences in WC, WHR, BMI, HDL and FPG of the genotypedistribution of TNF-α-308(P<0.05).Conclusion There were significant correlation among HSP70-1-110and TNF-α-308with metabolic syndrome in Ningxia; HSP70-1-110C allele and TNF-α-308A allele carriersincrease the risk of metabolic syndrome in Han and Hui people; The genotype and alleledistribution of HSP70-1-110,+190and TNF-α-308in Hui is consistent with the Han; Therewere significant correlation among C allele of HSP70-1-110with abdominal obesity withmetabolic syndrome, it carriers increased risk of metabolic syndrome; There were significantcorrelation between the level of occupational stress with metabolic syndrome in Ningxia, theodds ratios for MS increase with the level of occupational stress; The levels of occupationalstress were not related to the HSP70-1-110,+190and TNF-α-380gene polymorphism.There were significant correlation among TNF-α-308with obesity, hyperglycemia anddyslipidemia, it carriers increased risk of obesity, hyperglycemia and dyslipidemia.
