Aim: To investigate the expression of β-catenin、vascular endothelial growthfactor-C (VEGF-C)、P-glycoprotein(P-gp) in colorectal cancer (CRC) and to analyzetheir significance.Methods: The expression of β-catenin、VEGF-C and P-gp in50cases ofcolorectal carcinoma and14cases of para-cancerous tissues were detected by Envisiontwo-step immunohistochemical techniques, and then comprehensive analysis ofcombined the other clinical pathological factors such as patient’s age、sex、 tumor size、location、infiltration depth、degree of tumor differentiation、lymph node metastasis andDukes stage.SPSS17.0statistical software was used for statistical analysis, p-value lessthan0.05were considered significant. The expression of the three indicators and themajor clinical characteristics of CRC were analyzed by chi-square test.The correlationof the three indicators were analyzed by spearman rank correlation.Results: In normal colonic mucosa, mostly β-catenin was detected on themembrane of epithelial cells, and reduced membrane expression of β-catenin was notedin CRC. β-catenin showed cytoplasmic and nuclear expression in35.7%of cases ofnormal colonic mucosa compared with74%of cases of CRC,P<0.05. The positiverates of VEGF-C expression were significantly higher in tumor tissue than in normalmucosa (72%vs35.7%,P <0.05). The positive rates of P-gp expression had nosignificantly difference between tumor tissue and normal mucosa (78%vs71.4%,P>0.05).The expression of β-catenin had a significantly positive correlation with theexpression of VEGF-C and P-gp (r=0.555,P<0.05;r=0.531,P<0.05). The expression ofβ-catenin was not correlated with age、sex、 tumor size、location、degree of tumordifferentiation(P>0.05), but infiltration depth、lymphnode metastasis and Dukes stage(P<0.05). Conclusion:1. The expression of β-catenin was not correlated with age、sex、tumor size、location、degree of tumor differentiation, but infiltration depth、lymphnode metastasisand Dukes stage.β-catenin ectopic expression may infect the formation and themetastasis of colorectal cancer. β-catenin film express defects make Colorectal cancercells obtaining high proliferation activity and aggressive, and it could be one goodpredictor of judgment the invasion and metastasis in colorectal cancer.2. There were significant difference about the expression of VEGF-C in normalcolonic mucosa and CRC, and the VEGF-C positive expression rates were higher incancer tissue. VEGF-C may influence the formation and the progress of colorectalcancer by promoting the formation of lymphatic. We can look for the upstream ofexpression that lead to the expression of VEGF-C,and depress the signal pathexpression to realize the inhibition of cancer metastasis.3. The high expression of P-gp may cause the chemotherapy failed in CRC orthe early relapse, so look for the related pathways to inhibit the expression of P-gpprotein is particularly important, to realize the suppression in the formation of tumorand to improve the chemotherapy efficient and the patients,prognosis.4. The expression of β-catenin、VEGF-C and P-gp appeared positive or negativepathology of many common in cancer tissue and normal colonic mucosa.Cytoplasmicand nuclear β-catenin expression may induce VEGF-C and P-gp overexpression in CRC,which may play an important role in lymphnode metastasis and drug resistance inCRC.β-catenin is one of the most important Pathway proteins in CRC, and it play animportant role in the generation、the development and the invasion of CRC. β-catenin isexpected to become the new target for colorectal cancer treatment.
