BackgroundIn order to adapt to unfolded protein response caused by endoplasmic reticulumstress, the glucose regulated proteins refer to a set of proteins generated by cells. GRP78is an important member of the tribe. As molecular chaperones, GRP78’s mainphysiological function is involved in protein synthesis when the newborn polypeptidechain folding. The P21protein, as the product of the ras gene, can cause celldysfunction and sustained proliferation which lead to cancer. Recent studies show thatthe high expression of GRP and P21are found in many tumor cells, which are involvedclosely in the generation, progression and prognosis of carcinoma and supply newtheories for clinical diagnosis and treatment.ObjectiveTo study the expressions of GRP78and P21in infiltrating ductal breast carcinomaand adjacent tissues, to investigate the relationship and the clinical significance betweenthem and the clinical pathological features of infiltrating ductal breast carcinoma.Methods1.We collected35cases of infiltrating ductal breast carcinoma specimens and35cases of adjacent tissues (control group) by surgery. Specimens were confirmed bypathology and clinical data integrity. Within30minutes the surgical specimens in vitrowere quickly placed in frozen at-80℃,then the specimens were paraffined and sliced upinto3μm. Before the immunohistochemistry assay, review the pathological type of eachspecimen by HE staining.2.The immunohistochemistry assay was used for assaying the expressions ofGRP78and P21, and scored them under high-power microscopy. Analyze thecorrelation between the expression level of GRP78and P21and age, tumor size,histological grade, lymph node metastasis of breast carcinoma, etc with putting the data into SPSS17.0by χ2test and spearman correlation analysis. Level of significance wasset to0.05.Results1.In35cases of infiltrating ductal breast carcinoma,19cases (54.30%) werepositive expressions of GRP78; In35cases of adjacent tissues,3cases (8.60%) werepositive expressions of GRP78. GRP78expression in infiltrating ductal breastcarcinoma was significantly higher than adjacent tissues (χ2=9.66,P<0.01).2.In35cases of infiltrating ductal breast carcinoma,14cases (40.00%) werepositive expressions of P21; In35cases of adjacent tissues,1cases (2.90%) werepositive expressions of P21. P21expression in infiltrating ductal breast carcinoma wassignificantly higher than adjacent tissues (χ2=22.86,P<0.01).3.The levels of GRP78and P21expression were not correlated with age, tumorsize, histological grade, lymph node metastasis, ER, PR, C-erbB-2, moleculartyping,etc (P>0.05).4.In18cases of positive expression GRP78, the number of positive expressionP21cases was9(50.00%); In17cases of negative expression GRP78, the number ofpositive expression P21cases was5(29.40%). The consistence of GRP78and P21’sexpression was not found (r=0.21,P>0.05).Conclusions1. GRP78and P21were overexpressed in infiltrating ductal breast carcinoma,which guidance on the diagnosis and treatment of infiltrating ductal breast carcinoma.2. The levels of GRP78and P21expression were not correlated with age, tumorsize, histologyical grade, lymph node metastasis, ER, PR, C-erbB-2, molecular typing,etc.3. The positive expression rate of GRP78in triple negative breast cancers showedless than the non-negative trend, sample size can be expanded further in depth study.4. The expression of GRP78and P21in infiltrating ductal breast cancer was nostatistically significant correlation, indicating that no significant interaction between theexpression of GRP78and P21.
