Objective: The aim of this research is to study the effect of Enterococcus faecalislipoteichoic acid (LTA) on the proliferation activity and alkaline phosphataseexpression of MG-63cells.Methods:1μg/mlm,5μg/ml,10μg/ml,50μg/ml and100μg/ml E. faecalis LTAwere added to MG-63cell culture medium for3,12,24,48,72hours respectively. MTTwas used to detect the cell proliferation and alkaline phosphatase expression of MG-63.There were four different MG-63cellular density. Each group repeated three times.Results:1.Enterococcus faecalis LTA inhibited2×104/ml of MG-63cellssignificant strong compared with the other three cellular density.2×104/ml of MG-63cells was set to the standard density in the next study.2.To2×104/ml of MG-63cells, the inhibition effect of1μg/ml of E. faecalis LTAstarted from3h. The most strong inhibitory effect was100μg/ml of E. faecalis LTAused for24h.3.1μg/ml of E. faecalis LTA had already affected the expression of alkalinephosphatase at3h. The expression peak was detected when1μg/ml of E. faecalis LTAwas used for12h. the lowest one was the group which100μg/ml of E. faecalis LTAwas used for24h.The results of test of alkaline phosphatase showed that three hours’ application of1μg/ml of E. faecalis LTA on MG-63cells can influence the change of the expression ofalkaline phosphatase.12hours’ application of1μg/ml of E. faecalis LTA expressed thehighest level of alkaline phosphatase and24hours’ application of1μg/ml of E. faecalisLTA expressed the lowest level of alkaline phosphatase. Conclusions:1. E. faecalis LTA can inhibit the proliferation of MG-63cells.2. E. faecalis LTA can also inhibit the differentiation of MG-63cells whichdegrade the expression of alkaline phosphatase.3.The inhibition effect of E. faecalis LTA get stronger along with the time andconsistency changes.
