Objective:To study the expression level changes of NF-κB caused by E.f. LTA aswell as TLR2agonist to MG-63cells.Methods: MG-63cell was treated with10μg/ml Pam3CSK4(TLR2agonist) for48hours.Then100μg/ml E.f. LTA was added for24h and48h respectively.The control group was cultured as common way for24h and48h respectively. Sampleswhich were lysised by classic method were quantified by BCA assay.The proteinexpression level changes of NF-κB was detected by western blotting.The membraneswere scaned by using Bio-Rad image analysis software.GelPro was applied to analyzethe band.The NF-κB level refers to the comparison of the quantity of NF-κB band andthat of GAPDH band.Results:1.Compared to the control group,the experimental group had more NF-κBexpression when the MG-63cell was treated with E.f. LTA for24h(P<0.05).ButPam3CSK4didn’t play the effective role this time.There was no statistical differencebetween LTA/Pam3CSK4and LTA group.2.When the cell was treated for48h,the expression trend of NF-κB consistanted withthat of24h.The whole diference was Pam3CSK4affected the protein expression level.LTA/Pam3CSK4group showed higher NF-κB expression level than that of LTA alone.3.Compared to the results of24h group, LTA/Pam3CSK4group had more NF-κBat48h while LTA itself didn’t affected by the time difference.Conclusions:1. E.f. LTA can upgratulated the NF-κB expression level of MG-63cell. 2.The application of E.f. LTA and Pam3CSK4can upgratulated the NF-κBexpression level of MG-63cell.
