Objective:The puepose of the present study was to establish an early experimentalmodel of hematogenous metastasis in mice and discuss the feasibility of usingimmunoblotting for observe the tumour cells which were through hematogenousmetastasis.Methods: C57BL/6J mice were injected through the tail vein with stable infectedEGFP-positive B16melanoma cells(1×10~6cells/mouse) to establish an experimentalmodel of hematogenous metastasis. Then anaesthetized the mice, cut off their middleand inferior lobes of right lungs.Taking0.05grams of the them each to extract theprotein.And all these samples were analized by Western Blotting.Results: In vivo experiment,the mean values of signal strength got from the blotsof immunoblotting are50.264,30.473,11.742,5.315,3.625at2h,4h,8h,12h,24h afterintravenous injection. Showing a reduction of the process.While after24hours,thevalues of the blots are9.752,25.254,30.265,50.475, at3days,7days,10days and14days,which showing a raising progess.Conclusions:Western Blot,therefore,is a feasible method in the research on earlyprogess of hematogenous metastasis in mice. Immunoblotting can observe at least1×10~4tumour cells through hematogenous metastasis. As time went on, the growth oftumour cells through hematogenous metastasis were suppressed first and thenpromoted,which reflect a dynamic process in hematogenous metastasis.
