Objective:To investigate the role platelet GPIbα plays in the hematogenous metastasis of colorectal cancer and its mechanism.Methods:The platelet- poor plasma was obtained from the healthy volunteers and the colorectal cancer patients, the washed platelet was obtained from the healthy volunteers. Platelets were incubated with thrombin, colorectal cancer cells SW480 and other irritants. Get platelets from wide-type 6-week-old C57 mice, repeated the experiments and injected the colorectal cancer cells which incubated with platelets in a dosage of 1×10^7/mouse, observed the hematogenous metastasis of colorectal cancer cells after 2 weeks, 1month, 6months.The expression of GPIbα on the activied plalelets was detected by Western blot, realtime PCR and flow cytometry. Active paltelets were incubated with SW480, the conglutination of platelets and cancer cells was detected by flow cytometry and electron microscope.Results:1) The thrombin concentration in the plasma of colorectal cancer patients showed significant change compared with healthy volunteers.2) The conglutination of platelets and cancer cells showed significant difference, compared with the controls, which proved that colorectal cancer cells can update the conglutination of platelets and cancer cells.3) When injected the cancer cells which conglutinated with platelets, the hematogenous metastasis of colorectal cancer were more than the controls.4) After incubated with thrombin, the m RNA expression of platelet GPIbα, protein expression on membrane, and toltal protein expression showed significant difference.Conclusion:Platelet plays an important role in the hematogenous metastasis of colorectal cancer. There is a higher thrombin level in the colorectal cancer patient, it can lead to a higher expression of GPIbα on the platelet, subserved the conglutination of cancer cells and platelets, and finally cause a high incidence of hematogenous metastasis.
