Expression And Significance Of MMP-1TIMP-1in Liver Fibrosis Of Rats Infected With Paragonimusproliferus

[Objective] To investigate the dynamic changes of MMP-1, TIMP-1in rats infected with paragonimus proliferus, and to explore the roles of MMP-1, TIMP-1in Liver fibrosis formation.[Methods]25rats were randomly divided into two groups:the control group and infection group.There were5rats in the control group,20rats in the infection group.The animal model were set up by the way of peritoneal injection of8paragonimus proliferus cercarias to each rat in the infection group, while the equal amount of saline was injected to each rat in the control group. Infection group were divided into4groups according to infection time:at the3rd,5th,7th and9th week after infection,5rats each group. All rats were killed in corresponding time points after being infected. The levels of MMP-1, TIMP-1in the serum were determined by enzyme-linked immunosorbent assay. The liver tissues were sectioned and dyed by HE to determine the extent of alveolitis and liver fibrosis. The protein expression of MMP-1, TIMP-1in rat liver parenchyma was detected by immunohistochemistry.[Results] The results of HE staining:With prolonged infection time, the inflammation increased gradually in the liver tissue of rats, especially at the5th week, after liver cell degeneration necrosis increase gradually. The immunohistochemistry showed:With the progression of pathological changes, expression of MMP-1and TIMP-1both were increased gradually. Levels of MMP-1and TIMP-1detected by ELISA in the serum expression of MMP-1was increased,expression of TIMP-1was increased gradually. [Conclusion]paragonimus proliferus can cause liver fibrosis in rats. Expression of MMP-1and TIMP-1were different during the liver fibrosis formation. The extent of liver lesion was associated with the infection time and the expression of MMP-1, TIMP-1. MMP-1TIMP-1play a role in liver fibrosis of rats infected with paragonimus proliferus.
