Objective:Research on the effect of sea buckthorn seed oil on the expression of TIMP-1 in the liver of experimental rats with hepatic fibrosis. Methods: Randomly divide 80 healthy SD rats into 5 groups: sea buckthorn seed oil of low dose group, sea buckthorn seed oil of middle dose group, sea buckthorn seed oil of high dose group, normal control and model group.16 rats in each group.The exposure factors of each group are same(P<0.05). With CCl4 intraperitoneal set up the rat liver fibrosis model:to give other than the normal control group rats injected volume of 40% CCl4 pure peanut oil.The dose is 0.1ml/100 g,To H?M?S group rats lavage in sea buckthorn seed oil. The dose is 10ml/kg?5ml/kg?1ml/kg in turn,2 times a week,even need to 8 weeks. All treatments lasted for 8 week,then all rats were sacrificed.Take a portion of liver tissue to do pathological examination.Using the chart 2.2 standard to judge results.With BAD two-step detect the expression of TIMP-1 in liver tissue.Uses the ImageProplus(IPP) graphic analysis system to analyze optical density value. Calculate the mean optical density value and standard deviation in each group.Using one-way ANOVA to provide evidence that the difference was significant in the mean optical density of each group. Results: When at the end of 8 weeks experiment,14 rats dead. Mortality is 17.5%. Consider the cause of rats death is CCl4 poisoning.The pathological stage of the rest of rats was: in model group 8 rats was at 4-level, 3 was at 3-level; in H group 1 was at 3-level, 10 was at 2-level, 3 was at 1-level; in M group 2 was at 3-level, 11 was at 2-level, 1 was at 1-level; in S group 7 was at 4-level, 5 was at 3-level. Immunohistochemical results:the expression of TIMP-1 of model group(0.455±0.050)significantliy stronger than the normal control group(0.241±0.059)(p<O.05). The expression of TIMP-1 of H group(0.311±0.043)significantliy stronger than the normal control group,but weaker than model group,( p<O.05). Conclusion:Sea buckthorn seed oil has a certain inhibition effect on the expression of TIMP-1 gene in experimental liver fibrosis.
