Objective:To study the effects of the recombinant lentiviral vector for RNA interference(RNAi) of TLR4gene on the expression of TLR4gene and the growth ofsubcutaneous tumor of hepatocellular carcinoma in nude mice.Methods:Construct two miR-TLR4plasmid and a negative control plasmid, and select aplasmid which has obvious interference effect for lentivirus packaging. BALB/Cnude mice were inoculated subcutaneously with HepG3B cells to establish thesubcutaneous tumor model of human hepatocellular carcinoma. The mice wererandomly divided into experimental group, negative control group and blank controlgroup. TLR4miRNA lentiviral particles, lentiviral particles which carrying noeffective sequence or NS(0.2ml per time) were injected individually every two days.The volume of tumor of mice were measured. and the expression of TLR4wasdetected by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) and Western bolt.Results:After transfecting interference fragment1, TLR4protein expression quantity wassignificantly reduced, and the interference effect of interference fragment1was themost obvious (P<0.05). The mean rate of tumor growth in the experimental groupwas much slower than in the negative control group and blank control group(P<0.05).The dissected tumors in the blank control group and the negative control group hadweight of (2.32±0.03)g and(2.26±0.02)g, whereas in the experimental group, thetumors had a weight of (0.95±0.02)g, the tumor inhibition rate of experimental groupwas(58.12±0.35)%. And TLR4mRNA and protein in the experimental group weredecreased(P<0.05).Conclusion:TLR4miRNA lentiviral vector has a significantly inhibitory effect on the growthof subcutaneous tumor of hepatocellular carcinoma in nude mice.
