PART ONEObjective:To explore the tumor growth inhibition effect of docetaxel combined gamma knife on transplant subcutaneous hepatoma cells SMMC-7721in the nude mice.Method:To establish subcutaneous transplanted model of hepatoma in nude mice.The tumor-plant nude mice were randomly divided into2groups:Group1-Tumor control group without any processed;Group2-Docetaxel combined gamma knife irradiation group.The tumor-plant nude mice were processed in groups after30days to be detected the size and weight of transplanted tumors.The transplant subcutaneous hepatoma in the nude mice and different organs’morphology from each group were observed with microscopy in HE staining.Results:The tumor’s volume and weight in the group of using docetaxel were significantly less than control group and blank group (P<0.01).The group with Using docetaxel could decrease density of transplant subcutaneous hepatoma cells and varying degrees of the sheet necrosis was found in part of the hepatoma nests.Conclusion:Docetaxel could significantly inhibit the growth of transplant subcutaneous hepatoma cells SMMC-7721in the nude mice and promote the apoptosis of hepatoma cells to play the role of anti-hepatoma in vivo. Objective:To explore detection for proteins that are associated with cancer cells and apoptosis after the treatment with docetaxel combined gamma knife on transplant subcutaneous hepatoma cells SMMC-7721in the nude mice.Method:To establish subcutaneous transplanted model of hepatoma in nude mice.The tumor-plant nude mice were randomly divided into2groups:Group1-Tumor control group.Group2-Docetaxel combined gamma knife irradiation group.The tumor-plant nude mice were processed in groups after30days.The expression of p53、VEGF、Ki67、hTERT in each group of tumor tissues were detected by immunohistochemical method.The apoptosis in tumor cells was detected by end labeling (TUNEL)assay.Results:Group2-Docetaxel combined gamma knife irradiation group evidently reduced the expression of genes involved-p53、VEGF、hTERT、Ki67which played the role in cell apoptosis and proliferation(P<0.05).Conclusion:Docetaxel could significantly inhibit the growth of cells SMMC-7721in the nude mice.By regulating expression of antiapoptotic gene p53、hTERT,proliferous gene Ki67and angiogenesis inhibitive factors VEGF in transplant subcutaneous hepatoma tissue to promote the apoptosis of hepatoma cells.
