Objective To detect the protein and mRNA expression of T-box transcription2(TBX2), murine double minute2(MDM2) in tissues from breast invasive ductalcarcinoma, atypical ductal hyperplasia and normal breast tissues. To investigate theirroles in carcinogenesis, biological behavior and to evaluate their clinical significance.Methods The expression of TBX2and MDM2protein and mRNA in breast invasiveductal carcinoma tissues(n=90), atypical ductal hyperplasia(n=30) and normal breasttissues(n=30) were detected by using immunohistochemical SP method and in situhybridization method. Results1.The positive expression rates of TBX2and MDM2protein and mRNA in breast invasive ductal carcinoma were,respectively75.6%and81.1%,74.4%and76.7%. The positive rates of TBX2and MDM2protein andmRNA in breast invasive ductal carcinoma were significantly higher than that innormal breast tissues (P<0.01), The positive expression rates of TBX2and MDM2protein and mRNA in breast atypical ductal hyperplasia were respectively60%,63.3%,53.3%,56.7%. The positive rates of TBX2and MDM2protein andmRNA in breast atypical ductal hyperplasia were significantly higher than that innormal breast tissues (P<0.05,P<0.01).2.The expression level of TBX2wassignificantly correlated with tumor differentiation degree, higher TNM stage andaxillary lymph node metastasis(P <0.05). The expression level of TBX2mRNA wassignificantly correlated with higher TNM stage and axillary lymph node metastasis(P<0.05),but it was not related to the age and the tumor diameter(P>0.05).Positiveexpression of MDM2was related to the histological grade and axillary lymph nodemetastasis(P <0.05), Positive expression of MDM2mRNA was related to the histological grade (P <0.05),but it was not related to the age,size of tumor,(P>0.05).3.The expression of TBX2was directly correlated to that of MDM2(rs=0.240,P <0.05). The expression of TBX2mRNA was directly correlated to that ofMDM2mRNA(rs=0.285, P <0.01). The expression of TBX2and MDM2protein wasdirectly correlated to that of TBX2and MDM2mRNA(rs=0.285, P <0.01).Conclusion1.The expression of TBX2and MDM2was obviously upregulated inbreast carcinoma as well as atypical ductal hyperplasia, implying that was activated inthe early stage of breast carcinoma. They stimulated cell proliferation abnormally bythe way of avoiding cell death and joined in the whole process of the occurrence anddevelopment of breast carcinoma.2.The expression of TBX2and MDM2was usefulindex in breast carcinoma without lymphatics metastasis and well-differentiatedcompared to that in breast carcinoma with lymphatic metastasis andpoorly-differentiated, which promoted cell hyper proliferation and malignantphenotyoe transfomation. It was a useful marker in canceration and metastasis ofbreast cancer.3. The expression of TBX2and MDM2protein was directly correlated tothat of TBX2and MDM2mRNA,which suggests that have a coordinated effect in thecarcinogenesis and development in breast carcinoma. Abnormal expression of TBX2and MDM2is associated with the carcinogenesis and metastasis of breast invasiveductal carcinoma.Combined analysis of TBX2and MDM2may provid a theoreticalbasis for early diagnosis and evaluating prognosis of patients with breast cancer.
