Objective To establish an acute lung injury model by perfusing with seawater intotracheal intubation of rats.Methods One hundred and twenty eight healthy rats were randomly divided into fourgroups(each32), control group(CG: no any treatment) and three seawater perfusinggroup (SG1、SG2、SG4:the animal were given intratracheally1、2、4ml/kg of seawater,respectively.) The changes of semeiology were recorded after quantity sufficientseawater were infused into lungs of rats. For the rats in all groups, the respiratory rate,heart rate, arterial blood gases, the W/D ratio of pulmonary and their changes ofhistopathology were observed.Results Tachypnea, airway with white foam exudate, cyanosis and wet ralesappeared in the rats of seawater drowning groups immediately after perfusion ofseawater. Compared with control group, PaO2and PaCO2in seawater drowning groupswere significantly decreased (p<0.01). The low level of oxygenation index (<300mmHg) at30min time point in SG1, SG2groups, then gradually rising to mormal,and at each time point in SG4group. Comparison between each group, W/D ratio oflung tissue in SG4group is significantly higher than that of other groups (p<0.01).Pulmonary interstitial edema, alveolar septum fracture, red blood cells andinflammatory cells infiltration could be seen under macroscopy and light microscopein the pathologic changes of rat lungs. Conclution A stable rat model of seawater drowning induced acute lung injury can besuccessfully established by perfusion of seawater in4ml/kg dosage into rats. Objective To observe the effect of HIF-1α, ET-1, TNF-α, IL-6in the expression ofseawater drowning induced acute lung injury animal experimental process, discussedabove factor regulating effect of HIF-1α mechanism.Methods50rats were randomly divided into blank control group (control, group,CG group) and experimental group (seawater seawater group, group SG), copy ofseawater drowning induced acute lung injury animal model, according to the seawaterperfusion after0.5,1,2,4H is divided into SG1, SG2, time SG3, SG4group. TNF-α,IL-6content was detected in plasma ELISA, ET-1content in lung tissue of RIAdetection, IHC detection of HIF-α expression.Resutls compared with CG group, SG1, SG2, SG3, SG4groups, ET-1, HIF-1α TNF-α, IL-6content in the water after infusion of0.5h or higher, the difference wasstatistically significant (P <0.05), and the indexes of SG3group reached the peak.Conclutions SWD-ALI animal model group, the expression of HIF-1α and closelyrelated to inflammatory factor. ET through the vicious spiral interaction andinflammatory factors, exacerbation of lung injury, and thus indirectly up-regulated theexpression of HIF-1α, improve the tolerance to hypoxia state of body cells.
