The content of total flavonoids and chlorogenic acid in the flowers and leaves ofHuangshan tribute chrysanthemum collected in different growth periods were analyzed inthis thesis. In order to get the optimum extraction conditions, the extraction of totalflavoniods from C. morifolium flower and leaf were studied. Meanwhile, the phenoliccomponents of flower, leaf and stem in Huangshan tribute chrysanthemum(Chrysanthemum morifolium Ramat.) were analyzed. Through that we have got to thefollowing results:The content of total flavonoids was determined by UV-spectrophotometry and thecontent of chlorogenic acid was determined by HPLC. The results indicated that thecontent of total flavonoids and chlorogenic acid in flowers of Huangshan tributechrysanthemum was the highest in medium blossoming, and appeared large changes inpicking period. And the content of total flavonoids in chrysanthemum flowers was thehighest in interim picking period (125.41mg/g). However, it was the lowest in later pickingperiod (73.37mg/g). The content of chlorogenic acid in chrysanthemum flowers was inrange of4.92mg/g6.88mg/g. The content of total flavonoids and chlorogenic acid inleaves of Huangshan tribute chrysanthemum had rising trend during the growing time, andthe content of total flavonoids and chlorogenic acid was109.30mg/g and7.87mg/grespectively in the later growth stage.The method of ultrasonic assisted was considered to extract total flavonoids from C.morifolium flower and leaf. A single factor and response surface methodology wereconsidered to optimize the process condition. The best extraction rate of total flavoniodsfrom C. morifolium flower could be obtained with72%alcohol concentration at the rate oftwenty one parts of solvent to one part of dry flower with ultrasonic wave extracted for38minutes, the temperature of extraction at62. And the best extraction rate of totalflavoniods from C. morifolium leaf could be obtained with47%alcohol concentration atthe the rate of twenty four parts of solvent to one part of dry leaf with ultrasonic waveextracted for27minutes, the temperature of extraction at63.The phenolic components of flower, leaf and stem in Huangshan tributechrysanthemum were detected by HPLC-MS/MS. The results showed that thirteen kinds offlavonoids and nine kinds of phenolic acids were detected in the C.morifolium flower,eleven kinds of flavonoids and six kinds of phenolic acids compounds were detected in theC.morifolium leaf, and eight kinds of flavonoids and five kinds of phenolic acids compounds were detected in the C.morifolium stem. Meanwhile, it was interesting to notethat there were numerous compenents such as chlorogenic acid,1,5-Di-caffeoylquinicacid,3,5-Di-caffeoylquinicacid,4,5-Di-caffeoylquinicacid,apigenin-6-C-xylosyl-8-C-glucos-yl,apigenin-6-C-glucosyl-8-C-arabinosyl,apigenin-6-C-arabinosyl-8-C-glucosyl,luteolin-7-O-glucoside,luteolin-7-O-glucuronide,diosmetin-7-O-6”-malonylglucoside,andacacetin-7-O-6”-malonylglucoside that could be found in all flower, leaf and stem of C. morifolium,exceptfor the difference in concentrations. However, acacetin-7-O-galactoside was found in bothleaf and stem of C.morifolium but could not be detected in C. morifolium flower, andapigenin-7-O-glucuronide was only detected in the leaf of C. morifolium.
