Objective:To explore the expressions of Angiotensin Ⅱ and Angiotensin Ⅱ Type1Receptor in bladder carcinoma and normal bladder tissue. To analyze the relationship between Ang Ⅱ and AT1R expression and the clinical pathological features in bladder tumor. To evaluate their roles in carcinogenesis, invasion and metastasis as well as their clinical significances, to provide theoretical foundation for early finding and intervention and new target for targeted therapy bladder tumor.Methods:The expressions of Ang Ⅱ and AT1R at cell-level were detected by S-P immunohistochemical method in48cases of bladder carcinoma and normal bladder tissue. The expression of Ang Ⅱ and ATIR proteins were detected by western blot. The expression of ATIR mRNA were detected at transcription level by Real-time quantitative Polymerase Chain Reaction (qRT-PCR).Results:Immunohistochemistry showed that the positive expressions of Ang Ⅱ and ATIR in bladder carcinoma tissue were significantly higher than that in the normal bladder tissue (z=7.543,7.132,P<0.01). Their expressions were closely correlated with the histological differentiation and depth of invasion of the cancer was involved (z=2.843~4.285, P<0.05), but not associated with gender and age of the patients (z=0.346~0.011, P>0.05). There was a positive correlation between the expressions of Ang Ⅱ and AT1R in the cancer (r=0.681, P<0.01). From the point of view of the level of protein, The expression of Ang Ⅱ and AT1R in advanced bladder cancer was high, there was statistically significant(F=58.672、62.532, P<0.01).From the point of view of the level of mRNA, The expression of AT1R in advanced bladder cancer was high, there was statistically significant (F=55.422、66.682, P<0.01).Conclusion:Angiotensin Ⅱ and Angiotensin Ⅱ Type1Receptor play a role in the development of bladder tumor. The high Ang Ⅱ and AT1R expression promotes the invasion of the bladder tumor cell. Ang Ⅱ and AT1R have some correlations between the expression, suggesting that high expression of Ang Ⅱ and AT1R in cancer cell, may accelerate the evolution of bladder tumor.
