| Objective:Investigate the effect of C-reactive protein1059G/C genepolymorphism (CRP1059G/C rs1800947) associated with carotid atherosclerosisin patients which are Han population got cerebral infarction. Declare the riskfactors of atherosclerosis. In order to provide a theoretical basis for patients athigh risk for cerebral infarction were early effective prevention and treatment.Methods:1.Cases were randomly selected from Yueyang Hospital affiliated toHunan Normal University from January,2011to July,2012newly admittedpatients with cerebral infarction. The total cases is126. All patients werediagnosed by CT/MRI, after admission to general data acquisition and carotidartery ultrasonography. And on the next day of fasting and peripheral venousblood of5mL to complete the analysis of CRP1059G/C gene polymorphism.2.According to the results of carotid ultrasound examination, cerebral infarctionwere divided into non carotid atherosclerosis group and atherosclerosis group.According to the results of PCR,they were also divided into G/G, G/C group, todistinguish genotypes had different effects on the level of plasma C-reactiveprotein and blood lipids.3. Experimental methods and statistical processing:Usingmultiplex allele-specific polymerase chain reaction (ARMS-PCR) technique,using SPSS17statistical software package, count data between groups werecompared and using the chi-square test. Measurement data using mean andstandard deviation (X±S) said, compare with single factor and analyse variance.Analyse the differences in the distribution of CRP polymorphism of1059G/Cgene in between the two groups, using the chi-square test. Using logistic relatedrisk factors of carotid atherosclerosis in patients with cerebral infarction by regression analysis. There was statistical significance in P<0.05.Results:1. Non carotid atherosclerosis patients accounted for43.65%.2kindsof1059G/C genotype were detected (G/G, G/C), among the two groups of CRPgenotype1059G/C gene, the genotype G/G was more, non atherosclerosisgroup and atherosclerosis group frequency of G/G genotype was80%,93%.Among two groups of alleles Gã€C, the G alleles are the most common, nonatherosclerosis group and atherosclerosis group G allele frequency was90%,96.5%. By the chi-square analysis, frequency distribution difference between thetwo groups of alleles G, C and G/G genotype, G/C had statistical significance (P<0.05).2. Analyse the correlation between two groups of patients with cerebralinfarction C-reactive protein concentration and genotype, with statisticalsignificance (P<0.05).3.Analyse the influence factors of carotid atherosclerosisin patients with cerebral infarction: The analysis using the logistic regressionmethod, finally get the influencing factors are gender, history of hypertension,coronary heart disease, total cholesterol and CRP genotype of1059G/C gene.Conclusion:1. CRP1059G/C genotype G/G may be one of the risk factors ofatherosclerosis in patients with cerebral infarction process.2. Effect of CRP1059G/C polymorphism on atherosclerosis may be related to the level of plasmaC-reactive protein.3. The influencing factors of cerebral atherosclerosis are:gender, history of hypertension, coronary heart disease, total cholesterol and CRPgenotype of1059G/C gene. |
