Objective To analyze the possible microRNAs regulating LCRG1geneexpression.Methods Using miRNA target gene prediction software-PITA to predict the targetmicroRNAs of candidate tumor suppressor gene LCRG1; The expression levels ofmicroRNAs in29cases of laryngeal carcinoma and paired normal aryngeal tissuewere detected using Real-time PCR. According to results of bioinformatic analysis,luciferase reporter gene vectors of3’UTR of LCRG1gene were Constructed, dualluciferase activity were detected, preliminary analysis of the possible regulation ofexpression LCRG1by miRNAs.Results The number of miRNAs target candidate tumor suppressor gene LCRG1was649; According to results of the prediction, miR-21, miR-181d, miR-193b,miR-222, miR-1264were selected for further study, The expression of miR-21,miR-222, miR-1264were significantly increased in laryngeal carcinoma, comparedwith paired normal aryngeal tissue. luciferase reporter gene vectors of3’UTR ofLCRG1gene were successfully Constructed, miR-21, miR-181d, miR-1264cansignificantly decreased LCRG13’UTR luciferase activity.Conclusions miR-21, miR-181d, miR-1264may be involved in the Regulation ofexpression of candidate tumor suppressor gene LCRG1in laryngeal carcinoma.
