Objective: To observe the effects of the Ginger ginseng pinellia decoction on thereproductive toxicity and its pharmacological serum on the isolated smooth muscle.Moreover, its underlying action mechanism was elucidated.Methods:1.Reproductive toxicity experiment:Female mice were divided into two stages, respectively, the female mice wererandomly divided into4groups: blank control group, the ginger ginseng pinelliadecoction low-dose group, middle-dose group and high-dose group randomly. The firststage: Female mice were continuously fed withwith ginger ginseng pinelliadecoctionfrom14d prior to mating to the17th gestation day. The female mating rate,pregnancy rate and maternal visceral weight of pregnant mice were recorded. Thesecond stage The mice in the low-dose group, middle-dose group, and high-dose groupwere fed with dried ginger ginseng pinellia decoction from the16th day of gestationalto the17th day. The pregnancy mice were sacrificed at the sevnteenth day ofgestational, and the effects of the drugs on the body weight, embryonic developmentand the fetal appearance were observed.2Isolated smooth muscle experiment:Theactivities of isolated smooth muscle with Powerlab/8s biological signalswere observed and recorded. The impact of the serum containing of Ginger ginsengpinellia decoction on spontaneous activity of isolated smooth muscle and analysis was investigated and associated mechanism was elucidated.Results:1Reproductive toxicity experiment:Compared with blank control group, for female mice mating rate and pregnancyrate, there was no significant in the Ginger ginseng pinellia decoction each dosegroup(P>0.05);the growth rate of weight of the pregnancy mice in the low-dose group,middle-dose group, and high-dose group in the Ginger ginseng pinellia decoction groupwere markedly down regulated, but the rate of living, dead and absorbed fetus have nostatistically significant (P>0.05);and the fetal weight, visible part of fetal developmentdeformity and the teratogenic rate in high-dose grouphad no significant changes.(P>0.05).2Isolated smooth muscle experiment:2.1Compared with blank serum, the concentration of serum had inhibiting effecton the isolated uterine smooth muscle. They both reduced the tension,frequency anduterine activity of these muscles, and the differences were significant in highconcentration drug serum group(P<0.05or P<0.01). Compared with pituitrin group,the concentration of serum group had the inhibiting effect on the isolated uterinesmooth muscle tension and activity, and the difference was statistically significant(P<0.01).2.2Compared with blank serum, the concentration of serum had nhibiting effecton the isolated gastric smooth musle. They both reduced the frequency and uterineactivity of these muscles, and the differences were significant in high concentrationdrug serum group(.P<0.01).Compared with the ACh group, the concentration of serumgroup had the inhibiting effect on the isolated gastric smooth muscle tension, frequencyand activity, and the difference was statistically significant(P<0.01).Conclusion:The Ginger ginseng pinellia decoction in the low-dose group has no significanteffect on the female mice in reproductive function and the development of embryos,middle and high dose group have toxicity on maternal organs in pregnant rats, and the high-dose group of fetal rats teratogenicity certain。The Ginger ginseng pinellia decoction has the antiemetic effect, can inhibit uterinesmooth muscle contraction.
