Objective:Guided with the theory of Traditional Chinese Medicine (TCM) and combinedwith modern medical science study and experiment technique,using model rat as research object,this topic is to exposing the effect and mechanism of the granule of Ginseng and Radix Puerariae by observing the ultrastructure changes of diabetes rat retina,the level of the nitrogen oxide (NO) and the advanced glyeation end products(AGEs) in serum from the eye socketvein changes of diabetes rat retina.Thus,provide the theory foun-dation for clinicalapplication of the granule of Ginseng and Radix Puerariae.Methods:In the present study,we used a model of diabetic rat with alloxan(ALX)-induced.The diabetic model rats were randomly divided into control group(MG), the granule ofGinseng and Radix Puerariae group(SG)and doxium group(DG).Normal control group (NG)was also set.The Normal control groups and control groups were given drugs with normalsaline,the doxium groups and the Shenge granule groups were given drugs with Doxium andShenge granule.All rats were given drugs only once a day for6month.Measured bloodglucose (BG)、urine candy(UC)and body weight(BW)once half month.At the first、third andsixth month of the experimengts, two rats were randomly taked out from the everygoups,killed and collect speciments,watched indexs by means of pathomorphology,lightmicroscope,immuncytochemisty:(1) the level of NO and AGEs in serum from the eye socketvein changes of diabetes rat retina.(2) retina ultrastructura changes and treatment effects.Results:1.Survival of NG group is in good condition,the phenomenon of three more andone less’about the MG group is obvious.The phenomenon of three more and one less’aboutthe SG and DG group is less the MG group.2.The weight of NG group is rapid increased,and the weight of MG group is slow increased.DG group and SG group increased aftercure,It is obvious in SG group.3.The blood glucose and urine candy of MG group、DG groupand SG group are all the time higher than NG group(P<0.05),MG group’s blood glucoseand urine candy is the highest,The blood glucose and urine candy of SG group is thelowest.4.The average value of AGEs in MG group、DG group and SG group is obvious higher the NG group (P<0.05),MG group are all the time higher,SG group is lower the DGgroup (P<0.05),The comparison between groups were statistically significant(P<0.05).5. Theaverage value of NO in MG group、DG group and SG group is obvious lower the NG group(P<0.05),MG group are all the time lower,SG group is the most close to the DGgroup.6.Under electron microscope,we can see from the control group that:the neuron、neurofibril and glial filament et. The neuron has a complete structure and the neurofibriland glial filament are abundant and regulation.The apoptosis cell of neuron is increasegradually,and the number of neurofibril and glial filament is decrease gradually.thestructure disrangement of neurofibril and glial filament show an aggravating tendencygradually along with the course of1、3、6months in model group.The retina ultrastructuralchange,which rats are applied with the SG granule,is obviously better compared with thecontrol group.Conclusion: Adopt SG granule joins with the glycuresis model rat,the retina ultrastructuralchang is obviously better compared with the control group,and the SG granule is able toreduce glycuresis model rat serum middle AGEs contents,restrain NO damage and regulatingfunction in the whole.relieve the degree and development of DR,and show an satisfactoryeffect of preventing DR rat,which in early times.
