Objective:The purpose of this study was to detecte theexpression of Ki67and IGF-2in normal gastric tissue,atypicalhyperplasia tissues and gastric carcinoma.Investigate Ki67and IGF-2whether there is a correlation with the different sexes、 ages andpathological types of gastric cancer patients in their respective expressionand both of them.Methods: Using S-P immunohistochemical techniqueto detect the expression of ki67and IGF-2in different gastric tissues.Experimental sample: normal gastric mucosa (more than5cm out of thetumor cutting edge)20cases as a control group;20cases of atypicalhyperplasia tissues (10cases of mild dysplasia,10cases of severedysplasia);40cases in gastric carcinoma (15cases of adenocarcinoma,25cases of poorly differentiated adenocarcinoma) were advanced stage;80cases of experimental samples were taken from the Department ofPathology, Luzhou Medical College archived paraffin blocks fromOctober2010to October2011;the histological type of each specimen wasdiagnosised by two experienced pathologist.SP Immunohistochemicalwas used to detecte Ki67and IGF-2expression in the three grouptissues.To use the SPSS19.0experimental results for statisticalanalysis.Results:⑴In normal gastric mucosa, atypical hyperplasia tissues and gastric cancer tissues, the positive expression rates of Ki67were5%,35%,65%; positive rate in the nucleus of gastric cancer tissues (65%)was significantly higher than that in dysplasia tissues and normal gastricmucosa (35%,5%);the different expression rate of Ki67in gastric cancertissues,dysplasia tissues (x2=4.848, p<0.05)and normal gastric mucosa(x2=19.39, p <0.05)were statistically significant; positive expression rate ofKi67(35%) in atypical hyperplasia tissues was significantly higher thanthat in normal gastric mucosa(5%),and this difference was statisticallysignificant (x2=5.625, p <0.05); the positive expression of IGF-2ingastric cancer tissues, atypical hyperplasia tissues and normal gastricmucosa was62.5%,30%,5%;obviously, the different expression rate ofIGF-2in gastric cancer tissues,dysplasia tissues (x2=5.64, p <0.05)andnormal gastric mucosa(x2=17.95, p <0.05)were statistically significant;Inatypical hyperplasia the IGF-2positive expression rate (30%) was higherthan that in normal gastric mucosa(5%), difference was statisticallysignificant (x2=4.329, p <0.05);⑵the expression of Ki67in differentgastric cancer group were differences which are closely related to thedepth of invasion of gastric cancer tissue, degree of differentiation andlymph node metastasis (P <0.05),but it has nothing to do with tumor size,gender and age (p>0.05);the expression of IGF-2in different gastriccancer group were differences which are closely related to the depth ofinvasion of gastric cancer tissue, degree of differentiation and lymph node metastasis (P <0.05), but it has nothing to do with tumor size,gender and age (p>0.05);⑶Linear correlation analysis showed:Ki67andIGF-2expression in gastric carcinoma were positively correlated (r=0.406,p=0.009).(4)Multivariate binomial regression analysis ki67independently associated factors were:tumor size, depth of invasion andthe existence of lymph node metastasis;IGF-2was independentlyassociated with the existence of lymph node metastasis.Conclusion:1.The positive expression rate of Ki67in gastric carcinoma was higher thanthat in atypical hyperplasia tissues and normal gastric mucosa tissue,menwhil the expression of ki67in dysplasia is significantly higher than inthe normal gastric tissue; different expressions of ki67in gastric cancerwere closely related with depth of invasion, degree of tumordifferentiation and lymph node metastasis,but the Ki67has nothing to dowith the gender, age, tumor size.Prompting detection of the expression ofki67in gastric cancer is important,it can help us to judge the prognosis ofpatients.2.Positive expression rate of IGF-2in gastric carcinoma washigher than taht in atypical hyperplasia tissues and normal gastricmucosa,menwhil the expression of IGF-2in dysplasia is significantlyhigher than in the normal gastric tissue.Different expressions of IGF-2ingastric cancer were closely related with depth of invasion,degree oftumor differentiation and lymph node metastasis,but the IGF-2hasnothing to do with the gender, age, tumor size.It is reasoned that the detection of tissue expression of IGF-2can be used as an early diagnosisof judging gastric cancer, as well as one of the indicators of prognosis.
