Objective:To observed modified wu pi decoction on OHSS rat model of peripheral serumIL-6and E2, rat ovarian weight, abdominal capillary permeability and VEGF proteinexpression, and to explore modified wu pi decoction on OHSS intervention effect andmechanism, provide a theoretical basis for the prevention and treatment of OHSS.Methods:The choice of a minor female Wistar rats (SPF grade)60, were randomly dividedinto modified wu pi decoction group, indomethacin group, control group and blankgroup, n=15. Modeling Wistar rats for four consecutive days from22days to25days of age, subcutaneous injection of PMSG once a day, rats given subcutaneousinjection of hCG in26days (24hours after PMSG injection) once established OHSSmodel, subcutaneous injection at the same time give Jiawei Wuzi skin drinkindomethacin CMC solution, saline to27-day-old once a day. The blank group from22-day-old to26-day-old subcutaneous injection of saline once a day for fiveconsecutive days, while giving normal saline to27-day-old once a day.28-day-old(hCG or saline injection after48hours) anesthesia the rats in each group, eachweighing. Take five rats in each group were injected with1%Evan blue0.1ml,30min after along the abdominal midline abdominal incision, remove both ovariesweighed and collected peritoneal washings determination Evan blue OD value byspectrophotometer at700nm wavelength. Groups remaining10rats from the femoralvein blood, remove both ovaries in rats weighing ovarian tissue VEGF proteinexpression was measured by immunohistochemistry SP two-step; rats were detectedby ELISA and RIA peripheral serum IL-6and E2levels. Comparative analysis ofwhether there are differences between each set of data.Results:1. The control group,modified wu pi decoction group and indomethacin grouprat peritoneal fluid EB levels, ovarian weight, serum E2and IL-6level is higher thanthat of the blank group, there was statistically significant difference (P<0.05); modified wu pi decoction group and indomethacin group rat peritoneal fluid EBlevels, ovarian weight, serum E2and IL-6level.,modified wu pi decoction group andindomethacin group rat peritoneal fluid EB levels, ovarian weight, serum E2and IL-6level to be lower than that of the control group, there were significant differences(P<0.05).2. Dtected the expression of VEGF in the ovary of rats by immunohistoche-mistry, brownish yellow.It is expressed in rat luteal cells, granulosa cells. OvaryVEGF protein expression of rats in the blank group was significantly lower than thatin control group, modified wu pi decoction group, indomethacin group, withsignificant difference (P<0.01). Expression of VEGF protein in ovarian modified wupi decoction group and indomethacin group rats were significantly lower than those inthe model group, with significant difference (P<0.01).Conclusions:1. Modified wu pi decoction and indomethacin can effectively reduce OHSSmodel rat of ovarian weight, reduce rat peritoneal capillary permeability, play aninhibitory effect on ascites;2. Modified wu pi decoction and indomethacin can effectively reduce OHSSmodel rat peripheral blood IL-6, E2level;3. Modified wu pi decoction and indomethacin can effectively reduce theexpression of VEGF in OHSS model rat ovarian tissue;4. Modified wu pi decoction and indomethacin on model rats of OHSS is similarto prevention and treatment effect.
