Photodynamic therapy (PDT) is a clinically approved, minimally invasive therapeutic procedure that can exert a selective cytotoxic activity toward malignant cells. It is a two-step technique in which the delivery of a photosensitizing drug is followed by the irradiation of light. Activated photosensitizers transfer energy to molecular oxygen which results in the generation of reactive oxygen species which in turn cause cells apoptosis or necrosis. The use of nanoparticles as carriers of photosensitizers is a very promising approach because these nanomaterials can satisfy all the requirements for an ideal PDT agent.Objective:The experiment is to observe the inhibitory of Photosan-loaded hollow Silica Nanospheres (PS-loaded NSs)PDT on gastric cancer AGS cell under different experiment conditions and to primarily observe the mechanism of PDT.Methods:The human gastric carcinoma cell line AGS was selected to research, The experiment is to observe the inhibitory of Photosan-loaded hollow Silica Nanospheres (PS-loaded NSs)PDT on gastric cancer AGS cell under different experiment conditions and to primarily observe the mechanism of PDT. AGS were divided into six groups:group1was control cells receiving neither light nor drug; group2was light control for cells exposed to laser irradiation at a fluence; group3was drug control for cells incubated with free PS and not exposed to laser irradiation;group4was drug control for cells incubated with PS-loaded NSs and not exposed to laser irradiation; group5was cells receiving the experimental treatment with free PS and irradiation;while group6was cells receiving the experimental treatment with PS-loaded NSs and irradiation. The survival rate of cells was assessed by MTT method. And then the effect of PDT in the cell apoptosis were detected by flow cytometer(FCM).Results:The results of MTT method showed that both PS-loaded NSs and free PS had significant killing effect on AGS cell, The survival descended obviously(P<0.05)along with the PS-loaded NSs or free PS increased the concentration in some condition(P<0.05). FCM test showed PS-loaded NSs are more likely to cause cell apoptosis in the that same conditions.Conclusion:Compared with free PS, PS-loaded NSs can more effectively inhibit the proliferation of human gastric carcinoma cells, and more likely to cause apoptosis.
