we established an infectious abortion model of SD(Sprague Dawley)rats with lipopolysaccharide(LPS) and treated them with progesterone(P) and LPS antagonist LPS-RS to study the role of P and LPS-RS for fetus protection and the difference of expression of progesterone receptor(PR) and nuclear factor-kappa B(NF-kappa B) in the decidua of SD rats. So we can further discusse the mechanism of infectious abortion and the preventive and therapeutic effect of progesterone and LPS-RS.Methods:we put the males and females together and defined The day found the vaginal plug as the0day of pregnancy.All the rat was divided into5groups randomly. The first group was the control group. The rest of the four groups were the experimental groups, which were dealt with LPS (150micrograms per only) at the7th day of pregnancy. The rats of third group were treated with progesterone (2.5milligrams per only) at the6th day to11th day of pregnancy. The rats of forth group were treated with LPS-RS (250micrograms per only) at the7th day of pregnancy and1hour before treated with LPS. The rats of fifth group were treated with progesterone (2.5milligrams per only) at the6th day to11th day of pregnancy and LPS-RS (250micrograms per only) at the7th day of pregnancy and1hour before treated with LPS. We treated rats of each group as the following table. All the rats were killed at the13th day of pregnancy and the abdominal aortic blood and decidua were collected. We analyzed the rate of embryo resorption of SD rats and used the ELASA kit to test the level of progesterone in the serum. We also test the expression of PR and NF-kappa B in the deciduas of rats by western blot and analyzed the statistical differences of them.Result:1.The rats model of abortion was successful. The embryo resorption rate of the second group was higher than the first, the forth and the fifth group and there was statistical significance(p<0.05). The he embryo resorption rate of the third group was lower than the second group but higher than the first group, however there was no statistical significance(P>0.05). There was no statistical significance among the third,fourth and fifth group(p>0.05).2.we tested the progesterone level of serum and found that the second group was lower than the first, the third, the forth and the fifth group and there was statistical significance (p<0.05). There was no statistical significance among the third, forth and fifth group (P>0.05).3.We detected the progesterone receptor of decidua with Western Blot. The PR of the second group was lower than the first, he third, the forth and the fifth group and there was statistical significance (p<0.05). There was no statistical significance among the third, forth and fifth group (P>0.05).4. We also detected the NF-kappa B of decidua with Western Blot. The NF-kappa B of the second group was higher than the first and the fifth group and there was statistical significance (p<0.05) and there was no significant difference between the third, the forth group and the second group of NF-kappa B level(p>0.05) and there was no statistical significance among the third, forth and fifth group (P>0.05).Conclusion:1.LPS can increase the embryo resorption of SD rats and reduce the level of progesterone and the expression of PR and increase the expression of NF-kappa B in the deciduas.2.The progesterone can increase the level of progesterone and the expression of PR in the decidual.3.LPS-RS can go against the embryo resorption, increase the level of progesterone and the expression of PR and decrease the expression of NF-kappa B in the deciduas.
