Extracting Triterpenoids From Persimmon Stem,Purification Process And α-Glucosidase Inhibitory Preliminary Study

Diospyros kaki Thunb belongs to Diospyros L. and Ebenaceae family. Shidi is the drying calyx from the Persimmon which is also knowed as persimmon money, persimmon small, persimmons and persimmon calyx. Shidi,a traditional Chinese medicine herbs,mainly contains triterpenoids and tannins, which has been adopted in PharmacopoeiaThe paper put persimmon stem as test materials, using ultraviolet spectrophotometry-colorimetric detection methods, mainly studied extracting triterpenoids from persimmon stem, purification process and the extraction of persimmon stem on a-glucosidase inhibition. The results were as follows:1. The study investigated amount of two chromogenic agents and bath time when oleanolic acid was used as the standard sample. The results showed that it was the best way to use5%vanillin-acetic acid0.4ml, perchloric acid0.8ml, bath time20min. The content of triterpenoids was determined by UV-Visible Spectrophotometry. the regression equation of the standard curve is y=Y=12.8125X+0.0568(r2=0.9992).2. The test results showed that different varieties persimmon stem triterpenoid content in different harvest periods, May have the highest content of all harvested varieties in trite penoid. With the extension of growing seasons showed a downward trend, during the same period the triterpenoid content is quite different in different varieties, the triterpenoid content of san dai jin, shui shi and niu xin shi is higher in harvest period, the lowest ci lang content.3. Triterpenoids from persimmon calyx was extract with Ultrasonic Wave Extraction.The results showed that the optimum extraction was as follows:adding the rate of solid and liquid1:15,extracting30min,extating temperature50℃.4. The test draws that AB-8is more suited purified triterpenoids in Shidi by research four different types of resin static adsorption and desorption of triterpenoids in Shidi.To study the influencing factors of static adsorption process of extract with different resins. The results showed that the optimum extraction was as follows: Density of the extract liquor was0.8mg/mL, PH of the extract liquor was6.0, the column rate of the65%ethanol was at2BV/h,and the triterpenoids content of persimmon stem can reach40.49%.5.The study on α-glucosidase inhibitory activity from different extract parts of persimmon stem have shown that:N-butanol extraction areas and ethyl acetate extraction areas on inhibition activity of α-glucosidase have strong effect, petroleum ether and water-soluble parts of the activity is very low; aqueous extract of persimmon stem have strong inhibitory activity of α-glucosidase activity.
