Objective: To evalue the effect of FuJian tablet tonifying essence to produce marrowon the motor function impaired by cerebral ischemia and investigate its functionalmechanism relative to RhoA/ROCK2signal pathway.Methods:60cases meeting the inclusive criteria were selected and divided randomlyinto control group and treatment group,30cases in each group. All cases were givenconventional treatment with western medicine, and the cases in treatment group were alsogiven Fujian Tablets at the same time. After treatment for two months, the clinicalsymptoms, activities in daily life, myodynamia and muscular tension of the patients wereobserved; Electrocoagulation was used to set up MCAO rat model, and the rats weredivided into sham-operated group, model group and medicine group. Each group wasfurther divided into two sub-groups based on observation time, four weeks or five weeksafter operation.Each sub-group contined8rats. RT-PCR and Western-Blot were used todetermined the expression changes of RhoA/ROCK2genes and proteins of rats. Thecorrelation between these changes and BDA dye and GAP-43expression were analyzed.Results: Clinical studies showed that the improvement level of patients’ clinicalsymptoms, activities in daily life, myodynamia and muscular tension in the treatmentgroup was superior to that in the control group (P<0.05), and Fujian Tablets could improveobviously the motor function of patients in the recovery phase of cerebral ischemic strokewithout increasing the risk of taking more drugs; Experimental studies showed that at theend of the fourth week both the gene expression and the protein expression of RhoA and ROCK2in the model group were lower than those in the sham-operated group (P<0.05),and those in the medicine group was much lower. At the end of the fifth week, there was nosignificant difference between the gene expression or the protein expression of ROCK2inthe model group and those in the sham-operated group (P>0.05), while those in themedicine group was significantly lower than other two groups (P<0.01). Furthermore, atthe end of the fourth or fifth week after operation there was obviously negative correlationbetween the GAP-43expression in the medicine and the gene expression and the proteinexpression of ROCK2in the left cervical cord of the rats, only in the medicine group.Conclusion: Fujian Tablets could improve obviously the dyskinesia of the patientswith cerebral ischemic; Fujian Tablets could inhibit the signal pathway expression ofRhoA/ROCK in the cervical cord of uninjured side of MCAO rats, which may provide afavorable micro-environment for the remodeling of corticospinal tract and improveimpaired motor function.
