Objective: To investigate the effect of Fujian Tablet tonifying the liver and kidney on function of neurons-astrocytes-endothelium (N-A-Ec) after cerebral ischemic stroke and its role in neurological rehabilitation. Methods: Based on the theory of pan-brain, we developed middle cerebral artery occlusion (MCAO) rat model and observed the function changes of N-A-Ec in the brain. BWT was used to evalute the motor function of MCAO rats and LDP was used to measure remote cerebral blood flow (rCBF) in infact to reflect the changes of blood flow and funtion of neurons. To reflect states of axonal regeneration and rebuilding of synapse, immunohistochemical methods were used to observed the expression of GAP-43 and SYN; expressions of GFAP observed by immunohistochemical methods were used to reflect the activity changes of astrocytes and expression of P4502C11mRNA measured by RT-PCR were used to reflect indirectly the capability of astrocyte to secrete EETs which can regulate rCBF; linear correlation analysis was applied to analyze the correlation between GFAP and GAP-43 and between P4502C11mRNA and rCBF; in situ hybridization was used to observe the expressions of ecNOS, ET-1 and P-gpmRNA in endothelium to reflect the function of endothelim to regulate contraction-relaxation state of blood vessels and form blood-brain barrier (BBB). Results: Fujian Tablet could remarkably improve the impaired motor function and increase rCBF; as to neurons, it can increase the expressions of GAP-43 and SYN to promote the axonal regeneration and synaptic rebuilding; as to astrocytes, it can also increase expressions of GFAP, which indicates it can enhance the activity of astrocyte; the expression of P4502C11mRNA was also increased; the correlation analysis demonstrated that by administration of Fujian tablet the expression of GFAP was closely related to one of GAP-43 and the expression of P4502C11mRNA was closely related to rCBF; as to endothelial cells, it can inhibit pathological upregulated expression of ET-1mRNA andincrease expression of ecNOS and P-gp mRNA, which can be strengthened along with administration time. Conclusions: Fujian tablet with the effects of tonifying the liver and kidney can regulate the function of neurons, astrocytes and endothelium in vivo to increase rCBF, improve function of BBB and promote neurological structural reorganization, which may result in motor recovery in stroke.
