Lipoteichoic acid of Bifidobacterium can induce apoptosis of tumor cells in vitro and in vivo. But which mechanism inducing apoptosis is still unclear. Survivin is a member of the inhibitor of apoptosis family of protein (IAP), and plays an important role in the apoptosis proceeding, which can inhibit apoptosis induced by apoptotic stimuli such as TNF, Fas, VP16 and Taxol, etc. In order to explore the effect of LTA on Survivin in transcription and translation level, the expressions of Survivin in control and LoVo cells (colon cancer cells) treated with LTA were detected by RT-PCR and Western blot. Meanwhile, pAKT (the key protein kinase in PI3K/AKT signal transduction pathway), P53 and PTEN were measured by Western blot in order to illuminate the possible mechanism LTA regulating Survivin. We found there were overexpressions of Survivin mRNA and protein in control LoVo cells, while significant decrease of them in LoVo cells treated with LTA. Besides, the activity of AKT was decreased noticeably and the expressions of P53 and PTEN were increased, and they are dose-dependent.
