Object: To observe the synergistic effect of chemotherapy using Paclitaxol orLobaplatin combined with Trametes Robiniophila Murr and to study the influence ondrug sensitivity in human breast cancer MCF-7cell line and to explore the relatedmechanisms.Methods: This experiment uses human breast cancer MCF-7cell line as aobjective of study. Human breast cancer MCF-7cell line were treated with Paclitaxol orLobaplatin combined with Trametes Robiniophila Murr and mixture.Difference in drugresistance was analyzed between different regiments.1、MTT analyzed the growthinhibition by Trametes Robiniophila Murr, Paclitaxol and Lobaplatin in human breastcancer MCF-7cell line.2、Human breast cancer MCF-7cell line was treated bydifferent dose delivery of Paclitaxol and Lobaplatin, or combined with TrametesRobiniophila Murr, The growth inhibition were determined by MTT method.3、Humanbreast cancer MCF-7cell line was treated by Trametes Robiniophila Murr withpaclitaxel and lobaplatin, combination of two drugs as well as drugs soleadministration.Flow cytometer was used to detect the influence on cell cycle and cellapoptosis.4、 Human breast cancer MCF-7cell line was treated by TrametesRobiniophila Murr with paclitaxel and lobaplatin, combination of two drugs as well asdrugs sole administration.The number of colonies were determined through clonogenicsurvival assay, and then calculated the colony forming efficiency. Inverted microscopewas used to observe the influence on morphology of human breast cancer MCF-7cellby different methods of administration.Result:1、The results of MTT showed that Trametes Robiniophila Murr, paclitaxel and lobaplatin significantly inhibited human breast cancer MCF-7cell proliferation indoses-dependent ways;2、The results of MTT showed that Trametes Robiniophila Murr combined grouphad stronger inhibitory effects than paclitaxel and lobaplatin sole administration on thehuman breast cancer MCF-7cell.And there was statistical significance differencein the comparison between combined group and sole administration (P<0.05).3、The results of FCM showed that the apoptosis rate of medication groups allincreased and there was statistical significance difference(P<0.05). Apoptosis rate ofthe combined treatment group compared with the monotherapy group increased andthere was statistical significance difference(P<0.05).4、The cells ratio of the paclitaxel combined and monotherapy groups in G2/Mphase of MCF-7cell line was increased and S, G0/G1phase were decreased (P<0.05);cells ratio of the combined treatment group compared with the monotherapy group inG2/M phase increased and S, G0/G1phase were decreased (P<0.05). The cells ratio ofthe lobaplatin combined and monotherapy groups in G0/G1phase of MCF-7cell line wasincreased and S phase was decreased (P<0.05); cells ratio of the combined treatmentgroup compared with the monotherapy group in G2/M phase increased and S, G0/G1phase were decreased (P<0.05).5、Clonogenic survival assay showed that compared with control group treatmentgroups hadn’t cell colony formation,it prompted after treatment decreased cell colonyformation.Conclusion:1、Trametes Robiniophila Murr, paclitaxel and lobaplatin could inhibit the growthof human breast cancer MCF-7cell line.Inhibitory effect of the combined treatmentgroup compared with the monotherapy group is more obvious, it prompted TrametesRobiniophila Murr had sensitizing effect of chemotherapy on paclitaxel and lobaplatin.2.、Trametes Robiniophila Murr combined with paclitaxel and lobaplatin couldinhibit cell colony formation.3、Trametes Robiniophila Murr combined with paclitaxel induced cell cycle arrestat G2/M phase; Trametes Robiniophila Murr combined with lobaplatin induced cellcycle arrest at G0/G1phase.
