The Study Of Apoptosis Induced By Trametes Robiniophila Murr In Human Breast Cancer Cell Line MCF-7

Objective To study the growth inhibiting and apoptosis induced by Trametes robiniophila Murr in human breast cancer cell line MCF-7 and to approach its mechanisms.Methods To deal exponential phase of growth human breast cancer cell line MCF-7 with different density Trametes robiniophila Murr 48 hours,using MTT assay to detect OD value,calculating inhibition ratio.Cell morphology change was analyzed by optical microscopy and fluorescent microscopy .The induction of cell apoptosis by Trametes robiniophila Murr was analyzed by agarose gel electrophoresis of DNA ,flow cytometry. The protein expressions of p53 and p21 affected by Trametes robiniophila Murr were determined by Western blot. The expressions of p53 and Fas were determined by immunocytochemistry.Results The inhibition ratio of human breast cancer cell line MCF-7 increased with dentisy multiplication of Trametes robiniophila Murr.
