Abstract/Summary:
Objectives:1. Observe the state of breast cancer cell line proliferation after applying different concentration of Trametes Robiniophila Murr;2. Study the state of the side population cells of breast cancer cell lines after adopting Trametes Robiniophila Murr and epirubicin.Methods:1. Develop the breast cancer cell line, MCF-7and MDA-MB-231, in vitro from slow growth rate to rapid state of logarithmic phase;2. Apply MTT colorimetric method to observe the influence of the proliferation of human breast cancer cell line, MCF-7, and the MDA-MB-231, after adopting different concentration of Trametes Robiniophila Murr and epirubicin, each concentration (16.0,8.0,4.0,2.0,1.0mg/mL), and (4.0,2.0,1.0,0.5,0.25μg/mL);3. Apply flow cytometry analysis method of the side population cells to observe the side population cells of ratio state in breast cancer cell line MCF-7and MDA-MB-231, after using the medicine of Trametes Robiniophila Murr and epirubicin to that cells, each concentration4.0mg/mL and1.0μg/mLResults:1. MTT colorimetric method shows that the inhibition rate differences of Trametes Robiniophila Murr with different concentration to the two kinds of breast cancer cells has statistical significance (P<0.01);2. Trametes Robiniophila Murr, with different concentration4.0mg/mL,8.0mg/mL, and16.0mg/mL, bears the different inhibition ratio for the two kinds of cell lines, which plays a significant role in the statistic.(P<0.05);3. The proportion of side population cells of MCF-7cell line and MDA-MB-231cell line, each shows (1.70±0.20)%and (1.65±0.07)%, when we apply Trametes Robiniophila Murr, its concentration4.0mg/mL, to the two kinds of cell lines; the proportion of side population cells of MCF-7cell line and MDA-MB-231cell line each shows (3.49±0.10)%and (4.59±0.23)%, after using epirubicin, its concentration1.0μg/mL, to the two kinds of cell lines;4. Trametes Robiniophila Murr being used, the falling percent of side population cells in these two cell lines has a statistical significance.(P<0.05). Epirubicin being used, the falling percent of side population cells in these two cell lines has no statistical significance.(P>0.05)Conclusions:1. Trametes Robiniophila Murr significantly inhibits the cell proliferation of human breast cancer MCF-7and MDA-MB-231, with the concentration increased, the cell proliferation declines.2. The inhibitory effect of Trametes Robiniophila Murr with high concentration to the cell proliferation of MDA-MB-231is better than that of MCF-7cells;3. In the experimental concentrations, Trametes Robiniophila Murr is conductive to side population cells decreasing in two kinds of breast cancer cell lines, while, epirubicin fails to play a role in inhibiting the proliferation of the cells.
