Objective The experiment to detect the P2X7Receptor and vascular endothelialgrowth factor(VEGF) levels in vitreous fluid of patients with proliferativevitreoretinopathy (PVR), proliferative diabetic retinopathy (PDR) and normal control byenzyme-linked immunosorbent assay.To analyze the correlation between the purinergicreceptor P2X7and VEGF in proliferative vitreous retinopathy.Methods According to the patient’s general condition, eye-related history andfundus examination, the experimental group are as follows:①patients with proliferativevitreous retinopathy (PVR), a total of15cases (15eyes), male11cases, female4cases, aged35-72years old, mean age was48.3±10.6years old;②2-type diabetesmellitus patients accompanied proliferative diabetic retinopathy (PDR), a total of18cases (20eyes), male6cases, female12cases, aged48-75years old, mean age was64.3±7.2years old.③patients with idiopathic macular hole,10cases (10eyes), as anormal control group, male3cases, female7cases, aged46-71years old, mean age was59.3±12.1years old. A total of43cases(45eyes).There was no statistically differencebetween groups in terms of age, gender and so on (P>0.05).15patients in group PVRincluded B grade2cases,C grade10cases,D grade3cases (classification according to theretina standard classification society, published in1983), average history was27days(9-75days);18patients in group PDR included Ⅳ period8cases, Ⅴ period5cases,Ⅵ period5cases (7eyes),(installment standards with reference to the Chinesemedical association in1984diabetic retinopathy), with average12years (8-21years)history of diabetes mellitus, such as see things black shade, see things disappear, eyesshadow float and so on, ocular history with an average of5months (2-17months). Allpatients were strictly meet the vitreous cut surgical indications, before the surgery, signthe operation consent and sample collection informed consent,routine vitrectomy operation,put0.2-0.3ml intraoperative vitreous humor samples into1.5ml Eppendorffcentrifuge tube. Then the samples were preserved into-70℃refrigerator. After thesamples were collected complete by double antibody sandwich enzyme-linkedimmunosorbent assay (ELISA),to detecte the content of purinergic receptors P2X7andVEGF in the vitreous fluid of group PVR,group PDR, and normal control group.Anaylze the correlation between purinergic receptors P2X7and VEGF in group PVRand group PDR respectively.Results The content of purinergic receptors P2X7in the vitreous fluid of groupPVR was500.45±90.37pg/ml, while the content was369.72±116.24pg/ml in groupPDR. Both of them were significantly higher than the normal control group145.05±76.92pg/ml, the differences were statistically significant (P<0.01), the contentof purinergic receptors P2X7in group PVR was higher than that in group PDR, thedifference was statistically significant (P<0.05). The content of VEGF in the vitreousfluid of group PVR was249.64±36.90pg/ml, while the content was1289.69±295.77pg/ml in group PDR. Both of them were significantly higher than the normal controlgroup145.44±38.58pg/ml, the differences were statistically significant (P<0.01), thecontent of VEGF in group PVR was lower than that in group PDR, the difference wasstatistically significant (P<0.01).The purinergic receptors P2X7content and VEGFcontent were respectively positively correlated in the vitreous fluid of PVR group andPDR group.(R respectively.0.963,0.946, P <0.05).Conclusion1.The purinergic receptor P2X7content in vitreous fluid of PVRpatients increased significantly, and the increase amplitude is greater than VEGF,suggesting that P2X7may play a regulatory role in PVR occurrence and development,and its effect of prompted PVR progression is stronger than VEGF.2.The purinergic receptor P2X7content in vitreous fluid of PDR patients increased,and the increase amplitude is less than VEGF, suggesting that P2X7may be synergisticVEGF to play a regulatory role in PDR occurrence and development.3. PVR and PDR, the content of purinergic receptor P2X7and VEGF have somedegree of increased relative to the normal, suggesting that they in proliferativeretinopathy may play a certain role in promoting.
