Objective: Expression of Receptor for Activated Protein Kinase C1in manytumors included non-small cell lung carcinoma,atocellular carcinoma,breast cancer,oralsquamous cell carcinoma has increased, and it have certain relevance with the featuresof clinicopathological.The aim of the research is to detect the expression of of PTC,then analysis of the clinical pathological features and prognosis,and the correlation withKi67.Methods:we Collected thyroid related specimen of166cases confirmed to meetthe requirements during2005-2007from the department of Pathology of People’sHospital o f Liaoning Province, including76cases of PTC,30cases of NGWPH,30cases of nodular goiter,30cases of normal thyroid tissue. We detected the expression ofRACK1in specimens of136cases by using tissue microarray immune histochemicalmethod, analyzed whether RACK1has relevance with the clinical pathologicalcharacteristics of PTC or not. Moreover, Follow-up deadlines was January1,2012,follow-up time was0.9to60months, follow-up ending date was thyroid carcinomarecurrence or metastasis. We used the telephone follow-up. Survival time for patients istumor surgery day to diagnosis of recurrence or metastasis. Correlation analysis ofRACK1expression and the prognosis of the patientsResults: The positive expression rate of RACK1in normal thyroid tissue, nodulargoiter, NGWPH and PTC were26%,33%,33%,76%, respectively. Two comparisonbetween groups is shown that positive expression rate of RACK1in PTC has difference(P <0.05)with normal thyroid tissue, nodular goiter, NGWPH respectively; Betweenthe other groups (the nodular goiter and normal thyroid tissue, normal thyroid tissueand NGWPH, nodular goiter and NGWPH),there was no significant difference ofstatistics. The positive expression rate of Ki67in normal thyroid tissue, nodular goiter,NGWPH and PTC were13%,26%,33%,89%, respectively. Pairwise comparison results between groups showed that positive expression rate of Ki67in PTC has significantdifference (P <0.05). RACK1in76patients with PTC is related with tumor size,lymph node metastasis, clinical TNM staging (P <0.05). however, no correlation withthe patient’s gender, age.We also found that the positive expression of RACK1in76patients cases have significant correlation with positive expression of Ki67, with theenhancement of RACK1expressions,Ki67expression is also enhanced. The expressionof RACK1and Ki67has correlation, the correlation coefficient is0.301(P <0.05). Inaddition after surgical treatment of52cases of PTC patients were followed up for5years, we found that it is higher of RACK1positive recurrence after surgery patients,the difference was statistically significant (P <0.05).Conclusion: RACK1involved in a wide range of biological processes as ascaffolding protein, it was significantly higher in the expression of PTC, and itsexpression is associated with PTC TNM stage, Staging is higher, the better to express.Itcan be used as a papillary thyroid benign and malignant lesion identification referenceindicators combined with other factors. High expression of RACK1is closely relatedwith lymph node metastasis of PTC. Meanwhile, with the enhancement of Ki67expressions, RACK1expression is also enhanced. The high expression of RACK1canbe used as a independent prognostic marker of low survival rate of patients.RACK1may become the new therapeutic targets. It is likely to become the new treatmentstrategies if the expression of RACK1is suppressed in tumor cells,we can inhibit themetastasis and progress of tumor.
