The Eli VisionTM Plus immunohistochemical and in situ hybridization were used to detect the expression Snail, N-cad, E-cad in usual ductal hyperplasia, intraductal breast carcinoma and invasive ductal carcinoma.To investigate the expression of Snail, N-cad and E-cad protein and mRNA in invasive ductal carcinoma related to clinico-pathological features, the relationship among Snail, N-cad and E-cad protein and mRNA in invasive ductal carcinoma. In this study, two kinds of methods were adopted to analyze the consistency of Snail, N-cad and E-cad protein and mRNA by immunohistochemistry and in situ hybridization.The samples of tumors were taken from80cases of invasive ductal carcinoma,30cases of intraductal breast carcinoma and30cases of usual ductal hyperplasia, which were admitted to the251st Hospital of PLA from January2008to December2010. Respectively by means of EliVisionTM Plus immunohistochemical techniques and in situ molecular hybridization technical to detect the expression of Snail, N-cad and E-cad protein and mRNA. And to analyze the relationship of the age, tumor size, histological grade, lymph node metastasis, TNM stage, ER and PR expression in invasive ductal carcinoma with their relationship. Two kinds of methods were adopted to analyze the consistency of Snail, N-cad and E-cad protein and mRNA.The positive rates of Snail protein and mRNA were (13.3%,10.0%),(53.5%,46.7%),(82.5%,77.5%) in usual ductal hyperplasia, intraductal breast carcinoma and invasive ductal carcinoma, the expression of Snail among groups had statistical difference (P<0.05), the expressions of Snail protein and mRNA had no statistical difference (P>0.05). The expression of Snail protein and mRNA were related to lymph node metastasis, TNM stage and the expression ER, PR(P<0.05), but not related to the age, tumor size, histological grade (P>0.05). The positive rates of N-cad protein and mRNA were (10.0%,6.7%),(30.0%,20.0%),(60.0%,56.3%) in usual ductal hyperplasia, intraductal breast carcinoma and invasive ductal carcinoma, the expression of N-cad among groups had statistical difference (P<0.05), the expressions of N-cad protein and mRNA had no statistical difference (P>0.05). The expression of N-cad protein and mRNA were related to lymph node metastasis, TNM stage and the expression ER, PR (P<0.05), but not related to the age, tumor size, histological grade (P>0.05). The positive rates of E-cad protein and mRNA were (100.0%,93.3%),(73.3%,67.7%),(47.5%,42.5%) in usual ductal hyperplasia, intraductal breast carcinoma and invasive ductal carcinoma, the expression of E-cad among groups had statistical difference (P<0.05), the expressions of E-cad protein and mRNA had no statistical difference (P>0.05). The expression of E-cad protein and mRNA were related to histological grade, lymph node metastasis, TNM stage and the expression ER, PR (P<0.05), but not to related to the age, tumor size (P>0.05). The expressions of Snail, N-cad protein or mRNA in invasive ductal carcinoma showed positive relative relationship (r=0.363, P<0.01; r=0.430, P<0.01). The expressions of Snail, E-cad protein or mRNA in invasive ductal carcinoma showed negative relative relationship (r=-0.287, P<0.05; r=-0.324, P<0.01). The expressions of N-cad, E-cad protein or mRNA in invasive ductal carcinoma showed negative relative relationship (r=-0.654, P<0.01; r=-0.669, P<0.01).There were no statistical significance between Snail protein and mRNA or N-cad protein and mRNA or E-cad protein and mRNA, which demonstrated that there was no difference between protein and mRNA. Snail, N-cad and E-cad were involved in the occuring and development of breast cancer, and were related to lymph node metastasis, TNM stage and the expression ER, PR were regarded as a prognostic mark of breast cancer. The positive correlation between Snail and N-cad and the negative correlation between Snail and E-cad further demonstrated that they played an important role in breast cancer cell infiltration and metastasis.
